Objective:To explore the curative effect and mechanism of traditional Korean medicine Mahuangdingchuan decoction on allergic rhinitis in mice.Methods:Forty BALB/c female mice with specific pathogen-free were randomly divided into five groups:normal control group,allergic rhinitis model group,traditional Korean medicine Mahuangdingchuan decoction low dose group,traditional Korean medicine Mahuangdingchuan decoction high dose group,dexamethasone positive control group.Except for the normal control group,the other four groups were sensitized by intraperitoneal injection of 3%OVA every 3 days;At the end of sensitization,3%OVA was used for nasal drip and atomization inhalation for 8 consecutive days,The behavior of mice was observed immediately after each stimulation to determine whether the modeling was successful;The next day,the corresponding drugs were intragastrically administered to each treatment group for a consecutive week,changes in symptoms were evaluated after intragastric administration.In each of the above procedures,the normal control group was replaced with 0.3%sodium chloride in the same condiLions.Serum from nasal tissue and abdominal aorta of mice was taken the next day after excitation.The expressions of serum IL-4,IL-5,IL-13 and IFN-y were evaluated by ELISA,the histopathological changes of nasal mucosal tissue were observed by HE,the levels of NF-?Bp65 and p-p38MAPK in nasal mucosa and lung tissues of mice treated with Mahuangdingchuan decoction were detected by immunohistochemical staining and western blot.Results:1.The results of ELISA showed that the serum levels of IL-4,IL-5 and IL-13 in the rhinitis model group were higher than those in the normal control group,while the levels of IFN-y were lower(P<0.05).After the treatment with Mahuang dingchuan decoction and dexamethasone,the expressions of IL-4,IL-5 and IL-13 in serum decreased compared with that in the rhinitis model group,while the content of IFN-y increased significantly(P<0.05).2.No obvious glandular hyperplasia or other inflammatory cells were observed in the nasal mucosa of the normal control group by HE staining,and the nasal mucosa structure was intact.In the model group,there was abscission and necrosis of nasal mucosa with lymphocyte hyperplasia,more eosinophils infiltration in the submucosa and nasal mucosa edema.Compared with the rhinitis model group,the pathological morphological changes in each treatment group were less severe.3.The expression of NF-?Bp65 in the nasal mucosa of each group was observed by immunohistochemical staining.The results showed that the nasal mucosal structures of the normal control group were tinted less,without shedding,smooth and complete.The nasal mucosa of the rhinitis model group showed the epithelial cells,glandular cells and vascular endothelial cells were stained brown-yellow.Compared with normal control group,NF-?Bp65 protein content significantly increased.Compared with the rhinitis model group,the NF-?Bd65 protein content significantly decreased in the nasal mucosa of the Mahuangdingchuan decoction high and low dose groups and the dexamethasone positive control group.4.Westerm blot was used to detect the contents of p-p38 mapk and NF-?Bp65,The results showed that compared with the normal control group,the expressions of p-p38mapk and NF-?Bp65 in rhinitis mouse model group was higher significantly;the expressions intensity of p-p38mapk and NF-KBp65 in the Mahuangdingchuan decoction high and low dose groups and the dexamethasone positive control group were lower than that in the rhinitis model group.Conclusion:1.Traditional Korean medicine Mahuangdingchuan decoction is an effective medicine to relieve the local concurrence symptoms such as paroxysmal sneeze-like nasal obstruction and nasal itching caused by allergic rhinitis,can effectively inhibit the activation of NF-?Bp65 and the phosphorylation of p38MAPK.2.The key mechanism of Traditional Korean medicine Mahuang dingchuan decoction in treating allergic rhinitis is achieved by correcting the unbalanced Th1/Th2 immune network.
