The Effect Of Ornithogalum Caudatum Ait.on Proliferation And Apoptosis Of Human Lung Cancer H1299 Cells And Its Mechanisms

Objective Ornithogalum caudatum Ait.is a kind of a natural herbal plant,among which many of its chemical components have been shown to have potent antitumour activity.explore the effects of the extract of Ornithogalum caudatum Ait.on the proliferation and apoptosis of human non-small cell lung cancer H1299 cells,and to explore its mechanism。Methods H1299 cells were treated with different concentrations of the extract of Ornithogalum caudatum Ait.,Cell viability was measured by MTT at 24h、48h and 72h,After the treatment of H1299 cells with the extract of Ornithogalum caudatum Ait.,the morphology of the H1299 cells was observed by inverted microscope.After 48h of treatment with the extract of Ornithogalum caudatum Ait.;changes of apoptosis were observed by fluorescence microscope and the apoptosis rate was detected by flow cytometry.Expression changes of apoptosis-related proteins and gene bcl-2、Bax、Bad were detected by western blot and PCR。Results Compared with the control group,the inhibition rate of H1299 cells was increased after treatment by 0.5、1、2、5、10mg/mL on 48h and 72h(P<0.05、P<0.01);Under inverted microscope the typical morphological changes could be seen and the number of H1299 cells was reduced.Under fluorescence microscope the majority of the lung cancer H1299 cells were in late apoptosis,and in low concentrations group we can see few green fluorescent;Results of flow cytometry showed that the apoptosis rate was obviously dose-effect correlated with the concentration of the extract of Ornithogalum caudatum Ait.;Results of western blot and PCR indicated that compared with the control group,the protein and gene expression of Bcl-2 were down-regulated and the Bad was up-regulated,the rate of Bax/Bcl-2 were also up-regulated。Conclusions1.The extract of Ornithogalum caudatum Ait.can not only inhibit proliferation but also induce apoptosis of H1299 lung cells.2.The Bcl-2 family part of gene and protein also take part in induced apoptosis of H1299 lung cells which’ are induced by the extract of Ornithogalum caudatum Ait.
