Backgrand: RBMS3(RNA binding motif,single stranded interacting protein 3),a member of Myc single-strand bindingproteins(MSSPs)family,encoded RNA-binding protein.In mammals,the RBMS family consists of 3 members termed RBMS1-3.Recent studies shown RBMS3 have signifcant roles in tumor inhibition in some squamous cell carcinomas.However,the detailed function of RBMS3 in gastric cancer and its signifcance for clinical diagnose still remain unclear.Secreted frizzled-related protein(SFRP)family,which has 5 members(SFRP1-5).SFRP1 acts as Wnt /?-catenin antagonists and as inhibitor in numerous cancers.Aim: To detect the expression levels of RBMS3 and SFRP1 in human gastric cancer and corresponding normal tissues;to explore the relationship between the expression of RBMS3 and SFRP1 and the clinical parameters and prognosis of gastric cancer;to analyze the correlation between RBMS3 and SFRP1 in gastric cancer tissues.Methods: 23 pairs of fresh human gastric carcinoma tissues and corresponding adjacent tissues were collected for quantitative real-time PCR(q RT-PCR)and Western blot(WB)test;172 cases of gastric cancer tissues and 43 cases of adjacent tissues were used for immunohistochemical reaction.The expression levels of RBMS3 and SFRP1 m RNA and protein in 23 pairs fresh tissues were detected by q RT-PCR and WB,respectively.The expression of RBMS3 and SFRP1 proteins were further detected by immunohistochemistry in 172 gastric cancer tissues and 43 adjacent tissues.Results: Both RBMS3 and SFRP1 were expressed in gastric cancer and paracancerous tissues,but the expression levels of RBMS3 and SFRP1 in gastric cancer tissues were significantly lower than those in adjacent normal tissues(P<0.05).The expression levels of RBMS3 and SFRP1 in gastric cancer tissues were positively correlated.The expression levels of RBMS3 and SFRP1 in gastric carcinoma were positively correlated with the differentiation degree of gastric cancer(P<0.05);the expression level of RBMS3 was negatively correlated with the invasion depth of gastric cancer(P<0.05).In addition,the expression levels of RBMS3 and SFRP1 were positively correlated with the prognosis of gastric cancer patients(P<0.05),and the co-expression of RBMS3 and SFRP1 was an independent prognostic factor in gastric cancer(P <0.05).Conclusion: 1.RBMS3 and SFRP1 were highly expressed in gastric cancer tissues than those in adjacent tissues;2.The expression levels of RBMS3 and SFRP1 in gastric cancer tissues were positively correlated;3.low expression of RBMS3 and SFRP1 in gastric cancer indicated poor differentiation and poor prognosis;4.The co-expression of RBMS3 and SFRP1 may be a valuable molecular bio-marker for the prognosis of gastric cancer.
