Backgroud:In our country,the morbidity and mortality of lung cancer were the first in the malignant tumor,due to its easy local recurrence and distant metastasis,they influenced the health of the people.Chemotherapy drugs was the main purpose of killing cancer cells,but chemotherapy drugs killed normal cells in the body at the same time,lead to different degrees of cytopenia and thrombocytopenia.Thrombocytopenia of limiting the clinical dosage and application of the chemotherapy drugs,patients unable to tolerate further chemotherapy process,and make the treatment process forced to shorten or delay,or even stop,thereby seriously affecting the effects of the treatment of tumor and processes.However in recent years,studies have confirmed platelets promoted tumor growth and metastasis strongly,this may be caused one of the important reasons of bad prognosis of tumor patients.Platelets are a group of heterogeneous group,platelet parameters can not only reflect the change of platelet quality and quantity,but also reflect the platelet in the proliferation kinetics,so this study promotes a new evaluation system of tumor diagnosis and treatment between platelet parameters and tumor chemotherapy.Objective:To explore the relationship between platelet parameters and chemotherapy prognosis of patients with lung cancer.Methods:164 patients of lung cancer were retrospectively analysed.The changes of platelet parameters in lung cancer patients between 81 cases of the thrombocytosis and 83 cases of the non-thrombocytosis were compared.The differences of prognosis between two groups were also analysed.Results:The multivariate analysis with Cox hazard model showed platelet,TNM,pathology subtypes could be indicators for the prognosis of patients withlung cancer(P<0.05).There were no relationship between MPV,PLR,CEA,CY211,NSE and chemotherapy prognosis.The univariate analysis showed that the long-term overall survival have a statistic difference after chemotherapy in the two groups(P<0.05).Conclusion:Thrombocytosis is an independent risk factor for survival and MPV,PLR had no significant correlation with the prognosis of lung cancer chemotherapy,and Platelet may be a reliable indicator of prognosis evalulation of patients with lung cancer chemotherapy.Backgroud : Lung cancer is one of the most common malignant tumor of the clinical and its morbidity and mortality rate is on the rise.At present,the mortality rate of lung cancer has been the first.Infinite proliferation and metastasis are the significant reason of death in lung cancer,which also provides a conundrum for clinical treatment.As early as 1865,Trousseau has found the correlation between platelets and tumor.Malignant tumor is high coagulation state and common and rich in micro thrombus of platelets.Cancer cells grow without the restraints of feedback control mechanisms,leading to increased cancer cell survival.The treatment of cancer is often complicated by the lack of response to chemotherapy leading to chemoresistance and persistent survival of tumour cells.The previous clinical research we found that the platelet can affect the prognosis of patients with lung cancer chemotherapy.A growing number of studies reported also confirmed that platelets promoted the development of tumor.Therefore,this research explore the biology effects on lung adenocarcinoma in vitro environment by platelets and studied the role of platelets in chemotherapy-induced cancer cell death and survival,which to offer a new breakthrough point for early prevention and treatment of tumor.Objective:(1)To explore the influence of PLT in lung adenocarcinoma cell cancer proliferation and migration.(2)To study the role of PLT in chemotherapy-induced cancer cell.(3)Explore the mechanism of the platelet to lung adenocarcinoma.Methods:(1)Through CCK-8 experiments to observe the effects of A549 cell proliferation by different concentrations PLT.(2)By flow cytometry detected the apoptosis effect on A549 by PLT and the effect PLT to chemotherapy drug.(3)Through scratch test and Transwell experimental to observe the effects of migration ability of 549 by PLT.(4)Analysis the expression of ERK1/2,JNK,p-ERK,p-JNK,MMP 2,MMP-9,the Bcl-2,Bax after PLT and A549 cells co-cultured by western blot.Results:(1)Platelets can promote lung adenocarcinoma cancer cell proliferation,and presents the concentration dependence,in 48 h,proliferation effect is obvious;(2)By flow cytometry detected the apoptosis effect on A549 by platelet amd the effect platelet to chemotherapy drug.(3)scratch test and Transwell experiments have shown platelet enhance the migration ability of lung adenocarcinoma cancer cell;(4)Western blot have shown ERK1/2,JNK,p-ERK,p-JNK,MMP-2,MMP-9,Bcl-2 expression were significantly increased,the expression of Bax is decreased obviously in 48 h.Conclusions: Platelets stimulated growth and migration of A549 cells in vitro,which were positively correlated with platelet concentrations.Platelets stimulated the expression of ERK1/2,JNK,p-ERK,p-JNK,MMP-2,MMP-9 in MAPKs to promote proliferation and metastasis of lung adenocarcinoma.The resistance of tumor chemotherapy drug by Platelets showed that platelets may be a potential treatment targets for lung adenocarcinom combinating with chemotherapy drug.
