Purpose: Renal cell carcinoma(RCC),which has a higher degree of malignancy,is one of the most common malignant tumors in the urogenital system and the most common malignant tumor of the renal parenchyma.Because of absence of early obvious symptoms,signs and specific clinical manifestations in renal cell carcinoma patients,so it is more difficult to early diagnose.So that approximately one-third of patients with renal cell carcinoma has developed distant organ metastasis in times of the definitive diagnosis of the tumor.Since renal cell carcinoma is not sensitive to radiotherapy and general chemotherapy,surgical resection has become an effective treatment.However there are still 20-30% of RCC patients suffering tumor recurrence after renal carcinoma surgery.Therefore,it is highly significant to study the mechanism of the development of renal cell carcinoma and to find the molecular markers of early diagnosis,therapeutic evaluation and prognostic analysis of renal cell carcinoma.Micro RNAs(also known as miRNAs)are a class of short chain non-coding RNA molecules with a length of about 21-23 nucleotides,which regulate the expression of a wide variety of target genes at the post-transcriptional level though directly binding to some sequence-specifc sites of 3' untranslated region(3' UTR)of the target gene and following resulting in degration of the target gene m RNA or inhibition of it's translation.Recent studies have shown that miRNAs are differently expressive in a variety of tumor tissues,including renal cell carcinoma,and are involved in various life processes such as cell proliferation,invasion,migration,apoptosis,and so on.We took miR-216b-5p and renal cell carcinoma as the research objects to study the function and related carcinogenic mechanism of miR-216b-5p in the development and progression of renal cell carcinoma,which could provide sometheoretical basis for the further study of miRNA in the diagnosis and treatment of renal cell carcinoma.Methods: The expression of miR-216b-5p in 41 cases of renal cell carcinoma and adjacent normal tissues was detected by q RT-PCR.And the correlation between the expression level of miR-216b-5p in RCC tissues and the clinicopathological features of RCC patients was analyzed by statistical software.Then,the effects of miR-216b-5p on the RCC cell proliferation,migration and apoptosis were evaluated through CCK-8 proliferation experiment,cell scratch assay and flow cytometry respectively.Finally,the potential target genes of miR-216b-5p were predicted by bioinformatics methods.Results: The expression level of miR-216b-5p in renal cell carcinoma tissues was significantly lower than that in adjacent normal tissues(P < 0.05),and there was no significant correlation between the expression level of miR-216b-5p in RCC tissues and gender,age,pathological type,TNM stage and clinical stage of AJCC of the RCC patients(P > 0.05).After the expression of miR-216b-5p was up–regulated in RCC cells by transfecting synthetic miR-216b-5p mimics,the cell proliferation and migration of RCC cells were inhibited,and the cell apoptosis of them was promoted compared with the control group(P < 0.05).Conclusions: MiR-216b-5p is expressively down-regulated in renal cell carcinoma and plays an important role in tumor suppressor by inhibiting the cell proliferation and migration of renal cell carcinoma cells and promoting cell apoptosis of them.KRAS may be one of the target genes of miR-216b-5p.
