number of white blood cells and neutrophils;high sensitivity-C reactive protein;tumor necrosis factor-a;interleukin-6 and transforming growth factor-β1 expression in serum;Detection of serum total complement activity.Observation of histopathological changes in Rabbits with superficial soft tissue infection by Optical Microscopy.To observe the therapeutic effect and mechanism of hibiscus cream on superficial soft tissue infection in rabbits,and to provide theoretical basis for its clinical application.Methods:1 Bacteriostatic Experiment in vitro1.1 Grouping method:Concentration of 0.3g / ml Furong Paste powder solution for the experimental group,the choice of compound Huangbo paint and penicillin as a control group.1.2 Intervention method:Agrobacterium tumefaciens and Escherichia coli were used to inhibit the growth of Staphylococcus aureus and Escherichia coli.1.3 Observation index and detection method:It was observed that the three drugs could inhibit the growth of Staphylococcus aureus and Escherichia coli strains,such as inhibition,and the inhibition ring was recorded by the millimeter scale.2 In vivo experiment2.1 Grouping method50 New Zealand rabbits were numbered from low to high,and then divided into 5groups by random number table method.Each group was divided into blank group,model group,Baiduobang group,the high of Furong ointment group,the low of Furong ointment group,2.2 Modeling The back of the rabbits subcutaneous injection of Staphylococcus aureus 0.5ml(1.5 ×108CFU / ml),12 hours later see swelling and pain for the successful modeling.2.3 Intervention method Blank group: saline exudate hair removal,once a day,each rubbed about 5ml;Model group: saline outside the rubbed lesions,once a day,each rubbed about 5ml;Baiduobang group: model group,given more than 100 state coating,thickness 5mm,once a day;The high of Furong Ointment group: model group,given the thickness of the application of Furong Ointment 5mm,once a day;The low of Furong Ointment group: model group,given the thickness of the application of Furong Ointment 3mm,once a day.2.4 Observation index and detection method2.4.1 General growth:Naked eyes were observed in each group of rabbits after the daily use of pathological changes in tissue repair,eating,body hair gloss,limb activity and mental state,the use of electronic scales to measure the body weight of rabbits in each group after 7 days.2.4.2 Histopathological changes:The histopathological changes of rabbits with superficial soft tissue were observed by optical microscope.2.4.3 Inflammation(1)Blood routine:The number of white blood cells and neutrophils was measured by a blood cell analyzer.(2)Inflammatory factor:hs-CRP,TNF-a and IL-6 were treated with ELISA.2.4.4 Growth factor:TGF-β1 were treated with ELISA.2.4.5 Immune Function:CH50 also uses ELISA.2.5 Statistical methodAll data were treated with 17.0 SPSS statistical software package,P<0.05 that the difference was statistically significant.Measurement data using mean±standard deviation()that the group of rabbits after 7 days of weight measurement by group design data variance analysis;The number of neutrophils,neutrophil count,hs-CRP,TNF-a,IL-6,TGF-β1 and CH50 in the three time points were analyzed by variance analysis of repeated measurement design data.results1 Bacteriostatic test in vitro:The bacteriostatic ring of Hibiscus powder solution to Staphylococcus aureus was 2.5mm,there was no significant inhibitory effect on Escherichia coli.Compound Huangbai paint on Staphylococcus aureus and Escherichia coli were no significant inhibitory effect.Penicillin paper on Staphylococcus aureus antibacterial ring of 6mm,no significant inhibition of Escherichia coli.2 In vivo experimental results2.1 General growth:(1)During the experiment,there was no death in the normal group,the normal growth of body weight,no change in the skin of the hair removal area in the back,the normal eating,thick and shiny hair,good mental condition and brisk activity.No rabbits in the model group died during the experiment,body weight did not increase significantly,the back area red infection was still slow,repair,eating less,fur,fluffy and messy hair color less easy to fall off,gloss,listlessness,slow limbs activity.Baiduobang group,the high of Furong ointment group,the high of Furong ointment group of the above symptoms than those in model group decreased,but can not reach the level of normal control group.(2)Compared with the normal group,the weight of the model group was significantly reduced,P<0.01,indicating that the two groups were different;Compared with the model group,baiduobang group and the high of Furong ointment group and the low of Furong ointment rabbits’ in general growth were significantly improved,three groups of rabbits body weight increased significantly(P<0.01)2.2 Histopathological changes:Compared with the model group,the histopathologicalchanges of the back lesions of the Baiduobang group,the high dose group and the low dose group were obvious,the abscess was obviously reduced,the inflammatory cell infiltration was significantly reduced and the fibrous tissue hyperplasia was significant.Compared with the low dose group,the group of high dose group had less abscess,inflammatory cell infiltration,fibroblast proliferation and fibrous tissue proliferation.2.3 Blood routine change:Compared with the normal group,the white blood cells and neutrophils in the model group were higher than those in the normal group at three time points,P<0.01,with statistical significance.Compared with the model group,high dose group baiduobang Furong ointment group and low dose group of Furong ointment of rabbit whole blood leukocytes and neutrophils in three time points were significantly decreased(P<0.01).Compared with Baiduobang group and the high of Furong ointment groups in blood leukocytes decreased slightly,but P=0.333 was not statistically significant.Compared with the low of Furong ointment group and the high of Furong ointment groups in blood leukocytes decreased slightly,P<0.01,with statistical significance;compared with Baiduobang group at the three time points of blood neutral granulocyte in high dose group decreased slightly,P<0.01,with statistical significance.Compared with the high dose group,the whole blood neutrophils were high at three time points in the low dose group(P<0.05),which was statistically significant.2.4 Inflammatory factor changes:(1)Compared with the model group hs-CRP baiduobang group,high Furong ointment group and low ointment group of rabbits serum in three time points were decreased,respectively P<0.01;P<0.01;P=0.072,the low dose group was decreased,but the difference was not statistically significant.Compared with group hs-CRP high dose group serum baiduobang in three time points was slightly decreased,but P=0.985 was not statistically significant.Compared with the high dose group,the serum hs-CRP level in the low dose group was slightly higher than that in the high dose group,but only the mean difference was P=0.140,which was not statistically significant.(2)Compared with the model group TNF-a baiduobang group,high dose group and low dose group of rabbits serum in threetime points were decreased,respectively P<0.01;P <0.01;P<0.05,indicating that the three groups were statistically significant.Compared with the high dose group,the level of serum TNF-a in the high dose group decreased at different time points,P<0.05,which was statistically significant.Compared with the high dose group,the level of serum TNF-a in the low dose group was higher than that in the high dose group at different time points(P<0.01),which was statistically significant.(3)Compared with the model group,the levels of IL-6 in the serum of Baiduobang group,the high of Furong ointment group and the low of Furong ointment group were significantly decreased(P<0.01).Compared with the high of Furong ointment group,the serum IL-6 level was slightly lower at three time points,but P was 0.230,which was not statistically significant.Compared with the high of Furong ointment group,the level of serum IL-6in the low dose group was significantly higher than that in the high of Furong ointment(P<0.01),which was statistically significant.2.5 CH50 changes:Compared with the model group,the levels of CH50 in the serum of Baiduobang group,high dose group and low dose group were all increased at different time points,and the three groups were statistically significant(all P<0.01).Compared with Baiduobang group and high dose groups of rabbit serum CH50 three time points were slightly increased,but P=0.876 was not statistically significant.Compared with the high of Furong ointment group,the serum CH50 was significantly decreased in the low dose group at three time points(P<0.05),which was statistically significant.2.6 TGF-β1 changes:Compared with the model group,the levels of TGF-β1 in the serum of the Baiduobang group,the high of Furong ointment group and the low of Furong ointment group were all increased at the three time points,but only the model group and the high of Furong ointment group were statistically significant(P=0.090;P<0.05;P= 0.259).Compared with the high dose group,the levels of serum TGF-β1were higher in the three time points than those in the Baiduobang group,but only the mean difference was P=0.522,which was not statistically significant.Compared with the high of Furong ointment group,the serum levels of TGF-β1 were lower in the the lowof Furong ointment group at three time points,but P was 0.145,which was not statistically significant.Conclusion1 IFurong ointment inhibited the growth of Staphylococcus aureus.2 Furong ointment can regulate the levels of WBC,NE,hs-CRP,TNF-a and IL-6 in rabbit models,and has a certain anti-inflammatory effect.3 Furong ointment can regulate the expression level of serum CH50 in rabbits,and can enhance the immune function.4 Furong ointment can regulate the expression level of serum TGF-β1 in rabbits,and can accelerate the repair of diseased tissues. |
