Expression Of CXCL10,CXCR3 And T Cell Subsets In Peripheral Blood Of Advanced Vitiligo Patients,and The Influence Of Compound Chinese Medicine

Objective: Vitiligo is an autoimmune skin disease,and the disorder of the immune system leads to the occurrence of the disease.In this study,the chemokine CXCL10,CXCR3,CD3+CD4+ cells and CD3+CD8+ cells were measured,which were closely related to the immune system.The expression of CXCL10 and its receptor CXCR3 and T cell subsets were detected in peripheral blood of advanced vitiligo patients,as well as analyze the influence of compound traditional Chinese medicine.Methods: 28 patients with vitiligo and30 healthy controls were collected.The peripheral venous blood of patients and healthy controls were collected 10 ml.Flow cytometry was used to analyze T cell subsets,ELISA was employed to quantify CXCL10 and CXCR3,both in peripheral blood of non-segmental vitiligo patients before and after treatment and healthy control group.Results: The percentage of CD3+ CD4+ cells and CD3+ CD8+ cells in patients with vitiligo before treatment were lower than healthy control group but without significant difference(P> 0.05).The ratio of CD4+/ CD8 was higher but without significant difference either(P> 0.05).After treatment,the percentage of CD3+ CD4+ cells and CD3+ CD8+ cells were significantly increased in patients compared with the healthy control group(P <0.05).Before treatment,the expression level of CXCL10 is higher than the healthy control group(P <0.01).After treatment,CXCL10 decreased significantly(P <0.05).The expression level of CXCR3 was significantlyhigher than the healthy control group(P <0.05).There was no significant change in CXCR3 level after treatment.Conclusion Before treatment,the expression level of CXCL10 and CXCR3 were significantly higher than the healthy control group(P <0.01).Therefore,CXCL10 and CXCR3 are tightly related to the pathogenesis of vitiligo.After treatment,the expression of CXCL10 was significantly lower than before,so we can speculate that the compound Chinese medicine may be involved in the regulation process of immunity of patients with vitiligo.By decreasing the expression levels of chemokine CXCL10,thereby promoting the recovery of the disease.After treatment,the expression level of CXCR3 was not significantly higher than before,but it was still significantly higher than healthy control group.The results showed that the expression of CXCR3 was also correlated with the incidence of vitiligo.The proportion of T cell subsets was no significant difference between vitiligo patients and healthy controls,but after treatment,CD3+CD4+ and CD3+CD8+ cells proportion was significantly higher than before.The results showed that the use of compound traditional Chinese medicine has a regulatory effect on CD3+CD4+ and CD3+CD8+ cells.But due to the small number of cases,CD4(+)/CD8(+)had no significant difference between vitiligo patients and healthy controls,so further experimental studies are needed.In summary,CXCL10,CXCR3 and T cell subsets are involved in the pathogenesis of vitiligo.The compound traditional Chinese medicine used in this study can treat vitiligo by regulating T cell subsets,CXCL10 and CXCR3.
