Objective: By testing the expression level of chemokine receptor CXCR3 and chemokine ligand CXCL10 on ocular surface under different interventions.Analysis of the ocular surface of chemokines and their receptors expression characteristics and clinical features of dry eye. To explore the immune way to oprah ibuprofen eyedrops in the treatment of dry eye, To provide a scientific basis for prevention and treatment of dry eye. Methods: the Standard Patient Evaluation of Eye Dryness(SPEED) questionnaire shuould be finished Before and after medication on dry eyes.the same to normal control group.The tear film breakup time(BUT),corneal fluorescence staining(FLS) score and Sehirmer I test were performed in all patients.With Elisa method to detect the contents of TNF-α and CXCL10 in tears.Flow cytometry to detect T lymphocyte subsets in peripheral blood、 T lymphocyte infiltration in conjunctival epithelial cells and the expression level of CCR5 and CXCR3.The result of the three groups were compared. Results: The difference of the score of Standard Patient Evaluation of Eye Dryness(SPEED)questionnaire 、the tear film breakup time(BUT) and Sehirmer I test score of Dry eye people and normal controls has statistical significance(p <0.05).The difference of T lymphocyte subsets in Peripheral blood between dry eye and normal person has no statistical significance(p >0.05). The level of TNF-α、CD3 + 、CXCR3 and CCR5 of dry eyes is higher than normal(p < 0.05),But the difference of CXCL10 has no statistical significance(p >0.05).The expression of TNF-α、CXCL10、CD3+、CXCR3and CCR5 after treatment by only 0.1% sodium hyaluronate eye drops is lower than before,but the difference has no statistical significance(p >0.05).The expression of TNF-α、CD3+、CXCR3 and CCR5 after treatment by pranoprofen eye drops and 0.1%sodium hyaluronate eye drops is significantly lower than before(p < 0.05),and the differece of by CXCL10 has no statistical significance(p >0.05).(5)Positive correlationswere found in CXCR3、CCR5、CD3+ and TNF-αexpression with FLS and SPEED(p< 0.05)and a negative correlation in CXCR3、CCR5、CD3+ and TNF-α expression with BUT(p < 0.05) on 1 month in B group.Conclusion: Dry eye is a local inflammatory disease.Topical usage of pranoprofen eye drops combined with the 0.1% sodium hyaluronate eye drops Tis beneficial for remitting the ocular signs and symptoms in dry eyes.The possible mechanism action is pranoprofen can down—regulate the expression of inflammation factors and Chemokines in eonjunetival epithelial cells.
