Objective:To investigate the mechanism of Duhuojisheng decoction fumigation therapy curing the KOA by observing the changes of Notch3,Jagged1,and ACAN in the cartilages of machins with knee osteoarthritis.Methods: Part 1,forming the machin KOA-model: 6 machins were randomly divided into model group and blank control group,each group had 3.In the model group,the left knees were taken surgeries reference to the modified Hulth method.8 weeks later,all machins were put to death,the morphology changes of bilateral knees were observed,HE and Masson staining were accomplished for histologic examination,and scores were recorded according to the Mankin's score standard,in order to detect whether the KOA model is successful.Part 2,the influence of Notch3,Jagged1 and ACAN in cartilages of KOA models after the Duhuojisheng decoction fumigation therapy: After making sure the KOA model is successful,9 machins were randomly and equally divided into group A,group B and group C,all animals were made into KOA models.The group A were taken a Duhuojisheng decoction fumigation therapy combined with apparatus for 8 weeks,the group B were undergone a similar plan without apparatus,the group C were just used physiological saline for fumigation.After 8 weeks,the positive expression of Notch3,Jagged1 and ACAN in cartilage were observed and compared by immunohistochemistry.Result:(1)Visual observation: the peripheral connective tissues of left knees in model group had a obvious hyperplasia,synovial fluid was epinephelos,volume,and luteotestaceous;Cartilage surface was lack of luster associated with irregular ulcer or subchondral bone exposure.However,the synovium in blank control group was a little hyperplasia;synovial fluid was normal,liquid and flavescens;cartilage was holonomic without crack,erosion or ulcer.(2)The result of HE and Masson staining: The cartilage layer of KOA model became thin,broken,and had obvious crack,even worse presented coloboma and showed "zigzag" changes;cartilage cells significantly reduced,arranged disorder,cluster cells gathered.However,in normal cartilage,the four layer structures were clear,the tidal line was intact,cells arranged relatively uniform and had normal size and shape.(3)The outcomes of immunohistochemistry: The expression of Notch3,Jagged1 in group A was obviously inhibited compared with group B and group C;the expression of Notch3,Jagged1 in group B was significantly lower than that of group C,and there was significant difference in each group(P<0.05).But,The expression of ACAN in group A was obviously increased compared with group B and group C;the expression of ACAN in group B was significantly higher than that of group C,and there was significant difference in each group(P<0.05).Conclusion: Duhuojisheng decoction fumigation therapy could effectively inhibit the expression of Notch3,Jagged1 and the degradation of ACAN in machins KOA models,and combined with apparatus would have a better curative effect.
