Strychnos nux-vomica L.is toxiferous Chinese Tradition Medicine frequently used.It is well-known that Strychnos nux-vomica L and its prescriptions show excellent clinical therapeutic effect,especially in activating meridians to stop pain,and reducing swelling.But the dose of oral treatment and poisoning dose close to greatly limit its safe and effective clinical application.In recent years,with the development of transdermal drug delivery system,more and more Chinese herbal medicine and natural medicine have been developed into new products with definite dosage,lasting effect and less adverse reaction.Therefore,this article has carried on the system research of Strychnos nigra gel in detail,with a view to provide support for safe and effective application of Strychnos preparation.Mainly on the chemical composition of Strychnos,pharmacological toxicology of modem research progress and traditional Chinese medicine transdermal drug preparation technology evolution and modem research literature review.Which laid a theoretical foundation for the development of Strychnine Gels.In this paper,a new transdermal gel patch containing Strychnos nux-vomica L was developed and the prescription was screened and optimized by Box-Be-hnken response surface methodology.Herein,initial adhesion was tested for screening preparation conditions.The most optimized process route was predicted through Box-Behnken response surface method in optimizing the main factors.The experimental results exhibited that main excipient components of the gel patch was composed of sodium polyacrylate,polyvinyl pyrrolidone and Aluminium glycinate,and the most optimized ratio was 3.0(g):2.0(g):0.5(g)calculated by binomial equation with 0.8883 of multiple correlation coefficient r.Azone,piperitol and Tween-80 were screened for achieving expected transdermal results.The results showed that 3%Azone was superior to another two.The preliminary preparation technology of Strychnos nigra gel was studied.The results showed that the optimal parameters were 30min,and the coating was uniform in the early,middle and later stages.The transfer rates of brucine and strychnine from extract to finished product were 83.33%and 83.27%respectively,indicating that the process was feasible.The quality control method of the pilot product was studied.Determination of Strychnos nigra gel paste content of 21±1.5g/100cm2;adhesion force of the initial sticky force,the results can be stuck on the 11th ball;Strychnos paste placed on the tilt angle of 60? for 24 hours,no ointment surface flow phenomenon;excipient conform to pharmacopoeia requirements;content determination results for brucine content of 0.19mg/cm2,strychnine content of 0.11mg/cm2.The safety evaluation of the brucine gel paste was carried out.The acute toxicity,the single and multiple stimulation of rabbit skin and the allergic effect on skin of guinea pigs were evaluated.The results showed that Strychnos nujuba gel patch did not show any acute toxicity,irritation,skin allergies.The curative effect of brucine gel paste on bone cancer pain was evaluated.The effects of 60mg/kg,120mg/kg,240mg/kg of Strychnos nigra paste on rat bone cancer pain were studied in this paper.Finally,the changes of latency of mechanical stimulation were measured to determine the pain relief of Narcissus tazetta gel cream.There was extremely significant difference(P<0.01)between the high dose group 240mg/kg on the 14th day and the 16th day after the application of Strychnos nigra gel cream,and the claw reflex time of the model group,The gel ointment alleviated the hyperalgesic reaction of bone cancer pain model rats to a certain extent.
