Studying on HPLC fingerprints of Semen Strychni and Radix Glycyrrhizae has been described in this thesis. The main content is as follows:1.The HPLC fingerprint of Semen Strychni was eatablished. The extraction methods was ultrasonic for 30 min by methanol-H2O-HCL(50:50:1).Then determined by RP-HPLC(DAD), the chromatographic column was Kromasil KR100-5C18,flow rate at 1.0ml/min and the detection wavelength was 254nm.The gradient elution mode was applied in chromatographic separation,the mobile phase was acetonitrile(B)-0.2%triethylamine -0.2% glacial acetic acid water (A) (v/v): 0min,8%:92%;30min,17%:83%;60min,60%:40%.12 common peaks were found in the fingerprints of 10 batches of Semen Strychni,and good similarities with correlation coefficients higher than 0.99 were found in fingerprints between the herbs from different batches and the standard chromatographic fingerprint. The method is accurate and reproducible,providing a new reference for quality control of Semen Strychni.2.The HPLC fingerprint of Radix Glycyrrhizae was eatablished. The extraction methods is ultrasonic for 30 min by ethanol-H2O(50:50).Then determined by RP-HPLC(DAD),the chromatographic column was Kromasil KR100-5C18, flow rate at 1.0ml/min and the detection wavelength was 254nm.The gradient elution mode applied in chromatographic separation, the mobile phase was acetonitrile(B)- 1% glacial acetic acid water (A) (v/v):0 min, 13%:87%;30 min, 30%:70%;60 min, 56%:44%; 80min, 80%:20%; 90min, 80%:20%. 27common peaks were found in the fingerprints of 10 batches of Radix Glycyrrhizae, and good similarities with correlation coefficients higher than 0.93 were found in fingerprints between the herbs from different 9 batches and the standard chromatographic fingerprint. One batch was lower than 0.9. The method is accurate and reproducible, providing a new reference for quality control of Radix Glycyrrhizae.
