Study The Mechanism Of Semen Strychni Regulating The Immune Function On The Model Of Adjuvant Arthritis

Object: To observe the influnce of cell cytokines level which worked by semen strychni and study the mechanism initially.Methods: 40 healthy male rats were randomly dividided into 5 groups: normal group(Ⅰ),model group (Ⅱ), high dosage of semen strychni group(Ⅲ), low dosage of semen strychni group(Ⅳ)and methotrexate(MTX)group(Ⅴ).Inject the right rear-toe of theⅡ,Ⅲ,Ⅳ,Ⅴgroups with CFA to cause inflammation.After 18 hours ,when this model were successfully made,infuse steamed water 10ml·kg-1·d-1 from mouth in groupⅠandⅡ. GroupⅢwas given semen strychni 60 mg·kg-1·d-1, groupⅣwas given semen strychni 15 mg·kg-1·d-1 and groupⅤwas given MTX 0.5 mg·kg-1·w-1. After three weeks,detect the cytokines level of IL-1,IL-2,IL-6 and TNF- a from the rats'serum.Result:①The level of IL-1,IL-6,TNF- a in the model group is higher than other groups, the level of IL-2 in the model group is lower than other groups, Between groupⅡand other groups has a significant difference(P<0.05), but it has not significant difference with groupⅣ(P>0.05).②Between groupⅢand groupⅤ,the cytokines level of IL-1,IL-2,IL-6and TNF- a have no marked difference(P>0.05).③The level of IL-1,IL-2,IL-6,TNF- a of groupⅢhas a significant difference with groupⅣ(P<0.05).Conclusions: This subject proves that semen strychni can reduce the level of IL-1,IL-6,TNF-a from the rats'serum and improve the level of IL-2.The mechanism is probably that it regulate the immune disorder of RA.
