Objective: To explore the clinical significance of expression of miR-9 gene and Wnt/?-catenin in Xinjiang Uygur and Han with high-grade gliomas tissues.Methods:The expression of miR-9 and ?-catenin was examined by RT-PCR in formalin-fixed paraffinembedded primary tissue specimens in 49 cases from the First Affiliated Hospital of Xinjiang Medical University included 25 Han and 24 Uygur.In addition,2 epilepsy patients as control group with normal brain tissue.Analysising to its expression,accessing to 49 patients with clinical and pathological data,and patients were followed up.Then,we alse analyzed the relationship between the expression of these two makes and the clinicopathologic features of corresponding patients.The relationship between miR-9 and?-catenin mRNA expression and high-grade gliomas prognosis was evaluated using a Chi-square test,Cox regression model and Graph Pad Prism survival curve analysis.RESULTS: By RT-PCR there were 40,38 cases showed high expression of miR-9 and?-catenin,40/49(81.63%)?38/49(77.55%)respectively.The rates of positive mi R-9 and?-catenin RNA expression in high-grade gliomas were significant higher.The expression of miR-9 was positively correlated with that of ?-catenin(?2=12.382,P=0.002)by ?2 text.The expression of miR-9 and ?-catenin was not correlated with patient nationality,gender,location(P> 0.05).Single factor analysis determined that high expression of miR-9 and?-catenin,chemotherapy,radiation therapy and patient onset age were associated with high-grade gliomas patient prognosis(P<0.05).CONCLUSION: There were high expression in high-grade gliomas of miR-9 and ?-catenin.There was a positive correlation between the two indicators.Suggesting that miR-9 through the regulation of ?-cateninpathway affect the occurrence and development of high-grade gliomas.miR-9 and?-catenin plays a role in high-grade gliomas progression,and miR-9 and ?-catenin may be used as a prognostic marker in high-grade gliomas.
