Objective:Based on the previous study,we applied polar extract of cydonia oblonga and 3T3-L1 preadipocytes to reserach the effect and mechanism of polar extracts of Uyghur Cydonia oblonaga on improving insulin resistance in vitro.Methods:(1)Determine the best suitable cell density and drug concentration by MTT colorimetric assay.(2)Making 3T3-L1 induced into mature fat cells through the “cocktail” induction method based on references;the differentiation degree of adipoeyts was analyzed using spectrophotography quantitaviely after oil red Ostaing and detection of triglyceride.(3)The model of adipocyte insulin resistance was established by using culturing-inducing combination method of high-dose glucose and insulin;the change of glucose concentration in cell culture was determined after drug treatment.Result:3T3-L1 can be insulin resistance induced by cultured with dexamethasone.The model of insulin resistant is successfully established.The polar extract of uyghur Cydonia Oblonga can ameliorate the level of IR of 3T3-L1 at some point.And the mechanism maybe is upgrading the GLUT4 and IRS1 mRNA to ameliorate IR.Conclusion;The polar extract of uyghur Cydonia Oblonga can ameliorate the level of IR of 3T3-L1.And further studies are needed for clinical treatment. |
