Objectives:Using hyperlipemic rats, to observe the effects of total flavonoids ofCydonia Oblonga Mill (COM) on blood lipid metabolism and antioxidant activity, to explore the mechanism of the hypolipidemic effect. In addition,to study the effects of COM in different thrombosis model. Methods:1) Hyperlipemic rats established by high fat diet, with high serum total cholesterol (TC) concentration were randomly divided into control group, hyperlipidemia model group, quince leaves total flavonoids and quince fruit total flavonoid (40,80,160mg/kg) respectively in three groups, and simvastatin group.A fully automated biochemical analyzer was used to measure the following indicators:serum levels of cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), Alanine aminotransferase (ALT), aspartate aminotransferase (AST), Nitric Oxide (NO), Total Antioxidant Capacity (T-AOC) as well as liver concentrations of super oxide dismutase (SOD) malondialdehyde (MDA), glutathione peroxidase (GSH-PX), hepatic lipase (HL) lipoprotein lipase (LPL) after6weeks’treatment; and liver samples were observed for pathology.2) A study of the effects of total flavonoids of Cydonia Oblonga Mill on a mouse model of thrombosis by injecting collagen-adrenaline in the vein of the tail, on a rat model of arterio-venous bypass thrombosis and on thrombin-induced rabbit platelet aggregation in vitro. To observe the antithrombosis effect of total flavonoids of COM.Results:1) In induced hyperlipemic rats, COM total flavonoids significantly reduced serum TC, TG, LDL-C, ALT, AST and liver MDA (P<0.05or P<0.01), and significantly increased serum HDL-C, NO, T-AOC and liver LPL, HL, SOD, and GSH-PX activity (P<0.05).2) In antithrombotic experiments, Total flavonoids of COM significantly increased the survival of mice receiving a mixture of collagen-adrenaline; inhibited artery-vein bypass thrombosis and reduced thrombus weight in rats to different degrees.Plasma TXB2levels were reduced,6-keto-PGF1a was increased; inhibited rabbit platelet aggregation induced by thrombin. Conclusion:Cydonia oblonga has obviously adjusting blood lipid and anti-thrombosis effects.Quince fruit and leaf similar effect.
