Objective: The chemical analysis of Suhuang Zhike(SHZK)Capsule was studied,which will provide the chemical basis for the further antitussive constituents and action investigation.Method:(1)The essential oil samples of SHZK Capsule extracted by steam distillation and solvent extraction were tested using gas chromatography-mass spectrometry(GC-MS)with different injection methods,such as headspace sampling.The injection and preparation methods of essential oil samples were optimized and determined by the single factor and orthogonal experiments,and finally the best method was applied to the GC-MS analysis.(2)Different batches of essential oils in SHZK Capsule and their essential oil intermediates were analysized using GC-MS with the above condition,and the common chemical constituents of these two types of samples were investigated by the hierarchical clustering analysis and principal component analysis.(3)The analysis method of high performance liquid chromatography(HPLC)for non-volatile constituents was established,and primarily identified compounds by compared retention times with standard samples.(4)The amino acids of SHZK Capsule were derived using AccQ·Tag pre-column derivatization kit,and then also qualitatively analysized by compared HPLC retention times with amino acid standards.Results:(1)Compared with different sample injection and preparation methods,steam distillation is the best one for the essential oils of SHZK Capsule(i.e.add 10 times water,soak for 1.5 hours,extract for 7 hours with water vapour).The extract was analysized by GC-MS and identified 48 components,which comprised 64.59 %of the total essential oil.(2)The analysis results for different batches of essential oils in SHZK Capsule and their essential oil intermediates using GC-MS showed that there were no significant component differences during the actual manufacture of these two types of samples.(3)Compared HPLC retention times with standard samples,11 compounds and 14 amino acid compound were primarily analysized and identified.Conclusion: With GC-MS and HPLC,73 components in SHZK Capsule were analysized and identified,including 48 volatile components and 25 non-volatile components,which provided a comprehensive analysis of chemical constituents for SHZK Capsule.The analysis method is simple and repeatable,which will provide a good base for the further research of SHZK Capsule.
