This work intends to prepare Pt/Au/CuS nanoparticles using bovine serum albumin(BSA)as the biotemplate with evenly distributed size and favorable biocompatibility,followed by systematically exploring their potential applications in CT imaging,drug delivery and cancer photothermal therapy.The dissertation includes three composites:1.Albumin-mediated platinum(Pt)nanocrystals for in vivo enhanced computedtomography(CT)imaging.A simple albumin-directed fabrication of platinum(Pt)nanocrystals is presented for the utilization in CT imaging.We then systematically invested the applications of the nano contrast agents for in vivo mouse organs reinforced imaging such as heart after intravenous tail injection.It is found that the nanocryatals could serve as potential new and potent CT contrast agents,especially vital for in vivo imaging with prominent enhancement and metabolizable behaviours.2.Albumin-mediated gold(Au)nanoparticles loaded gemcitabine for cancercomputed tomography(CT)imaging and therapy.Gemcitabine(Gem)loaded Au@BSA nanospheres are fabricated for the efficient delivery of therapeutic agents into the A549 cells and simultaneous computed tomography(CT)imaging.The resulting positive charged Au@BSA nanospheres were conjugated with the negative Gem by electrostatic layer-by-layer assembling interactions.Our results demonstrated that the obtained biomimetic Au@BSA nanospheres were biocompatible with low cytotoxicity while the Gem-loaded ones exhibited obvious effects with efficiencies significantly greater than that of the free Gem.3.Albumin-mediated copper sulfate nanoparticles(CuS)for cancer photothermal therapy.Herein,a simple albumin-directed fabrication of copper sulfate(CuS)nanoparticles is presented for the utilization in cancer photothermal therapy.The in vitro results show that the CuS@BSA nanoparticles exhibit effient killing capabilities for A549 cells under laser irradiation(980 nm).Besides,they could also be utilized as new T1 contrasting agents for magnetic resonance imaging(MRI)applications.
