Objective: To research the conditions of expression of TSC1 in urothelial carcinoma of bladder(BUC)tissue, and analyze the relationship with clinical stage, pathological grade and recurrence. Methods: The expression status of TSC1 in 51 cases of urothelial carcinoma of bladder tissues and 5 specimens of normal bladder tissues which were choosed by immunohistochemical technique, and analyse its relationship between clinical stage, pathological grade and recurrence.Result: They all express of TSC1 in 5 specimens of normal bladder tissues, but the positive expression rate of TSC1 in bladder cancer was39.2%,and with the increase of tumor stage and pathology grade, the negative expression level of TSC1 become getting higher and higher. The negative expression rate of TSC1 in non-muscle invasive bladder cancer was 52.4%,and the negative expression level of TSC1 muscle invasive bladder cancer was 88.9%.Respectively,there exist statistical significance(52.4%vs88.9%,p<0.05).The negative expression rate of TSC1 in low grade urothelial carcinoma of bladder was44.4%,and in high grade urothelial carcinoma of bladder was 75.0%.Respectively,there exist statistical differences,(44.4%%vs75.0%,p<0.05).In this study,we followed up 45 bladder cancer patients for 12-36 months, and among which 19 bladder cancer patients were recurred after operation. The negative expression rate of TSC1 in recurrence group and non-recurrence group was 73.7% and 38.5%.Respectively,there exist statistical significance between recurrence group and non-recurrence group(73.7%%vs38.5%,p<0.05).In this study,we also found there were no relationship of TSC1 expression with sex and age of patients, and the size of tumor. Conclusions: The negative expression level of TSC1 in urothelial carcinoma of bladder tissue was higher than its negative expression degree in normal bladder tissue, and there exist relationship between TSC1 with clinical stage, pathological grade and recurrence. This raise the interesting possibility that the negative expression of TSC1 can contribute to the development of urothelial carcinoma of bladder. This may demonstrate that TSC1 can serve as markers to prognosis and estimate the progression of urothelial carcinoma of bladder.
