Background and Objecktive Cell cycle refers to all the process from the begining of the end of a metosis,which includs G1,S,G2 and M state.The length of the cell cycle may change itself beyond the difference of cells,but it is mainly determined by the state G1,because the others are usually fixed.The research of cell cycle is widely used in all kinds of tumors pathogenesis,providing increasing significance of the diagnosis,treatment,development and progressing of the tumors.Condyloma acuminatum,CA,which has the characteristic of strong infectivity,fast growth and easy to relapse after treatment,is one of the sexual transmitted diseases and it is common in dermatology clinic.CA is caused by human papilloma virus(HPV)and patients with CA always complain about symptoms,such as the genital verrucous thickening of skin and mucous membrane.The treatment ways of CA is mountainous,the traditional therapeutic methods,such as sugery,liquid nitrogen refrigeration and CO2 laser therapy,etc,can remove the wart body,but,always following with other problems which lead to unsatisfied trestment results.5-aminolevulinic acid photodynamic therapy,ALA-PDT,which is a new method can cure many disease including CA.5-aminolevulinic acid has high affinity of sick tissue,which can produce iradiation toxicity and destroy the abnomal tissues targetly under the certain intensity of light source.This kind of treatment method has lower recurrence rate and only makes small trauma,so it is aplide widely in dermotology clinic to cure CA,but,now,the precise mechanism of the ALA-PDT is still lack of systematic research,especially the effection of ALA-PDT on cell cycle before and after treatment has not been reported from now on.The study mainly talks about the effection of ALA-PDT on cyclin D1 and Cyclin E through the change of the expression intensity and positive rate before and after ALA-PDT,as to further improve the mechanism.Materials and Methods 1 Materials 42 patients of CA came from the Dermatology & Venereology Clinic of the First Affiliated Hospital of Zhengzhou University,which includes 23 males and 19 females.And the age of the patients rages from 17 to 65 years old,with the average age of 34.15 years old;The course of disease was from 20 to 100 days long,and the average of the age is 50.46 days;all the clinical manifestations of the patients were typical,acetic acid white test is positive,at the same time the histopathological diagnosis was definite,the in situ hybridization confirm that all the patients were infected with HPV,all the cases meet the clinical and laboratory diagnostic criteria of CA.All the patients had no any other treatments before the ALA-PDT and had no other systematic diseases.2 Methods The ALT-PDT treatment was composed to the 42 patients.Before the treatment,the wart body were taken under local asthmia by sugery as the control group.One week later,at the same palce of the wart body another wart tissues were taken as the experimental group.The immunohistochemical SP method were applied to detect the positive expression of Cyclin D1 and Cyclin E of the CA tissues respectively.Results determination:The positive expression of Cyclin D1 is mainly in the nuclear of princkle cell layer,and the Cyclin E is mainly in the shallow layer of the epithelium,such as the granular layer and the prickle layer.All of them are in the nuclears,which is full of brown granules and the boundaries are clear.Semi-quantitative grading was performed according to staining intensity and the percentage of the positive cells in the CA keratinocytes.Staining intensity scoring: not stained,pale yellow,yellow and brown yellow just for 0 point,1 point,:2 points and 3 points;Percentage of positive cells to total cells:0,1,2,3,4point respectively refers ?10%,11%~25%,26%~50%,51%~75%,?76%.multiplying the final score of the two scorings obtain the final score.0,1-4,6-8,9-12 respectively represents weakly positive(+),positive(++),negativestrongly positive(+++).Statistical treatment In my study all the data was statistically analiesed by the SPSS17.0 statistical software.The difference Cyclin D1 and Cyclin E positive expression rates between the two groups before and after the ALT-PDT treatment was compared with ?2 test;The differences in expression strength was tested by rank-sum test.the significant level =0.05,P<0.05 for the difference was statistically significantResults 1 The expression of Cyclin D1 before and after the treatment of ALA-PDT The positive coloration of Cyclin D1 of CA is mainly located in the nucleus,with clear nucleus and border brown particles.Among the42 cases,23 cases shows the positive expression of Cyclin D1,the positive expression rate is 54.76%(23/42),expression strength mostly at--~++;And there were only 6 cases of the patients showing positive expression,and the positive expression rate is 14.26%(6/42),the expression strength mostly at-~+ after the treatment,And the difference of positive expression rate of Cyclin D1 was statistically significant between the two groups before and after the treatment(?2=15.50,P<0.05);At the same time,the difference of positive expression strength of the two groups was statistically significant before and after the treatment(Z=-3.09,P<0.05).2 The expression of Cyclin E before and after the treatment of ALA-PDT The positive coloration of Cyclin E is mainly located in the nucleus of CA,with clear nucleus and border brown particles.Among the42 cases,36 cases shows the positive expression of Cyclin E,the positive expression rate is 85.71%(36/42),expression strength mostly at ++~+++;And there were 25 cases of the patients showing positive expression,with the positive expression rate of 59.52%(25/42),and expression strength mostly at-~++ after the treatment.The difference of positive expression rate of Cyclin E between the two groups before and after the treatment was statistically significant(?2=7.24,P<0.05);At the same time the difference of positive expression strength of the two groups before and after the treatment was statistically significant(Z=-3.032,P<0.05).Conclusions 1.The expression rate and the expression strength of Cyclin D1 is lower after ALA-PDT compared with before,it may implies that the ALA-PDT could inhibit the expression of Cyclin D1.2.The expression rate and the expression strength of Cyclin E is lower after ALA-PDT compared with before,it may implies that the ALA-PDT could inhibit the expression of Cyclin E.3.The inhibition of ALA-PDT on Cyclin D1 and Cyclin E which will inhibite the G1/S phase transforation of cell cycle and finally restain the cutin cell proliferation maybe one of the mechanism of how ALA-PDT treat CA.
