| Objective:To study the effect of PDGF/BMP in promoting bone repair around implants.Methods:24 rabbits(weighing about 2.2 kg)were implanted into the iliac crest to made implant animal models.The rabbits were divided into 4 groups randomly and injected diffent drug around the implants: 0.9% NaCl group,PDGF-BB group,BMP-2 group,PDGF-BB+BMP-2 group.In 3,7,10 days,the animals were killed,and then we took out the tissues around the implants.With Masson staining to check fibrosis and by alizarin red staining to observe calcification.Result:1.Masson staining:(1)0.9% NaCl group showed that the organization of collagen fibers appearred in 3 days,and produced abundant collagen fibers in 7 days,10 days;(2)PDGF-BB group showed that more collagen fibers than the NaCl group in 3 days,and then the fiber repair capacity was increaseing in 7 days,10 days;(3)BMP-2 group showed that the collagen fibers were visible less than the NaCl group in 3 days,but the collagen fibers and repair ability were increasing in 7,10 days;(4)BMP-2 +PDGF-BB group showed that the collagen fibers were visible less than the NaCl group,but the collagen fibers and repair ability was increasing in 7,10 days,and the repair ability was stronger than BMP-2 group.Alizarin red staining:(1)0.9% NaCl group showed no change in calcium deposition;(2)PDGF-BB group showed that the deposition of calcium salt decreased with the prolongation of time;(3)The BMP-2 group showed that the deposition of calcium salt decreased with the prolongation of time,but the effect was lower than that of PDGF-BB group;(4)BMP-2 + PDGF-BB group showed that the calcium salt deposits were somewhat reduced around the implant.Conclusions:1.The effect of PDGF-BB was stronger than that of BMP-2 in promoting bone healing around implants in the application of single factor;2.PDGF-BB+BMP-2 group showed more collagen fibers appeared in the early stage,and emergence of more calcium salt deposition later,and then shortening the bone integration process after implantation. |
