ObjectiveThis study adopts the method of flow cytometry,Prospectie study the changes of peripheral blood lymphocyte subsets ratio of early and oligonucleotides metastases of lung cancer patients before and after SBRT.Explore impact of SBRT for early non-small cell lung cancer and oligonucleotides metastases on the immune function.Materials and MethodsFrom October 2015 to January 2017,total 22 early non-small cell lung cancer and oligonucleotides metastases patients in Henan province tumor hospital radiotherapy department received SBRT treatmen, including 18 male cases, female 4 cases. The median age 74 years old(48-84).14 cases of stage I lung cancer,8 cases of metastasis.Collected peripheral blood 0 to 7 days before radiotherapy and 0 to 7days,4 weeks after radiotherapy.Applying flow cytometry to lymphocyte subgroup detection: CD3 +(total T lymphocytes),CD3 +CD4 + T(helper T cells),CD3 + CD8+(cytotoxic T cells),CD4 + CD25 +(regulatory T lymphocytes,Treg),and CD8 +CD28-(suppressive T lymphocytes,Ts),and CD8 + CD28 +(cytotoxic T lymphocytes,Tc),CD4 + / CD8 +,(CD4 + and CD8 + ratio).Using SPSS22.0software for statistical analysis, categorical variable described by the frequency and percentage.Continuous variables described by the mean±standard deviation.Paired t test is used to contrast the change of lymphocyte subsets before and after the treatment itself. The statistical differences(P < 0.05).ResultTotoal 22 patients collected peripheral blood 0 to 7 days before radiotherapy and 0to 7 days after radiotherapy to lymphocyte subgroup detection. After radiotherapy 0 to7 days,22 patients CD3+ increases than before radiotherapy(P=0.023).Other lymphocyte subsets ratio change is not obvious(P>0.05).11 patients collected peripheral blood four weeks after radiotherapy to lymphocyte subgroup detection, the lymphocyte subsets ratio change is not obvious than 0 to 7 days before radiotherapy.13 early stage lung cancer patients lymphocyte subsets ratio change is not obvious between 0 to 7 days before radiotherapy and 0 to 7 days after radiotherapy(P>0.05).9 oligonucleotides metastases patients CD3+(P=0.027),CD3+CD4+(P=0.038)and CD4+CD25+(P=0.032)increases than before radiotherapy.Other lymphocyte subsets ratio change is not obvious between 0 to 7 days before radiotherapy and 0 to 7 days after radiotherapy(P>0.05)ConclusionSBRT treatment have little influence on the immune function of patients with lung option for patients with medically inoperable early stage non-small cell lung cancer(NSCLC)and oligonucleotides metastases patients. It is a safe and effective treatment. In addition.SBRT treatment may cause tumor local immune response, the specific biomarker detection, detecting method, detecting timing needs further research.
