Objective Through the intervention of Yiqi Jianpi Tongbian in rats with slow transit constipation,the effects of the spleen time,intestinal propulsion rate and colonic myoelectric activity on the rats were observed,which was used for clinical application of Qi and spleen.The prescription lays a theoretical foundation for the treatment of slow transit constipation.Methods 1 modeling and grouping methods The STC rat model was prepared by intragastric administration of compound diphenoxylate(10 mg.kg-1.d-1).95 healthy clean Wister rats were divided into two groups for modeling.After successful modeling,STC model rats were randomly divided into model group,Yiqi Jianpi Tongbian low-dose group,Yiqi Jianpi-tong.The middle dose group,Yiqi Jianpi Tongbian high dose group.2 groups of intervention methods The low,middle and high doses of Yiqi Jianpi Tongbian were administered by Yiqi Jianpi Tongbian with daily doses of 6g/kg,12g/kg and 18g/kg,respectively,2ml each;normal group,model group Rats were intragastrically administered with 10 ml/kg of normal saline daily.The stomach was administered daily for 4 weeks.3 observation indicators and detection methods 3.1 Effects of Yiqi Jianpi Tongbian Recipe on the number of feces and water content in rats with slow transit constipation: Collect feces 24 hours after intervention in each group,record the number of fecal particles,observe the traits,and weigh the electronic analytical balance.The water content was measured.3.2 The effect of Yiqi Jianpi Tongbian Recipe on the discharge time of the first black stool in rats with slow transit constipation: After establishing the STC rat model,after giving Yiqi Jianpi Tongbian Recipe,it was given 10% activated carbon suspension.After the stomach,the time was counted with the activated carbon suspension,and the time of the first black stool was recorded for each rat.3.3 The effect of Yiqi Jianpi Tongbian Recipe on the intestinal propulsion rate of rats with slow transit constipation: The rats were sacrificed after 10% activated charcoal suspension,and the intestines at the lower end of the pylorus were taken out to the end of the rectum.In the state,the length of the pylorus to the rectum and the length of the activated carbon suspension in the intestine are measured,and the rate of advancement of the carbon in the intestine is calculated.3.4 Effect of Yiqi Jianpi Tongbian Recipe on colonic electrophysiological activity in rats with slow transit constipation: The anesthetized rats were placed on the constant temperature operating table on the supine position,and the lower abdomen was cut longitudinally after disinfection to expose the ileocecal area.A pair of electrodes were placed at the proximal end of the colon about 2 cm from the cecum,and the lead was connected to the BL-420 S biological function experimental system through the incision to measure the myoelectric activity of the colon in rats.4 statistical methods All measurement data were expressed as mean±standard deviation(x±s).The data of the two groups were compared by single factor(AVOVA)analysis of variance and treated with software SPSS 22.0.P<0.05 was considered statistically significant.Results 1 Comparison of fecal pellets and fecal water content: the number of fecal pellets in the normal group was(41.6±5.8),the number of fecal pellets in the model group was(27.8±8.6),and the number of fecal pellets in the low-dose group of Chinese medicine was(33.7±4.1).The number of fecal pellets in the middle dose group of Chinese medicine was(36.1±2.9),the number of fecal pellets in the high dose group of Chinese medicine was(39.1±4.0),the moisture content of feces in the normal group was(58.7±2.8)%,and the moisture content of feces in the model group.(45.9±7.4)%,the water content of feces in the low dose group of Chinese medicine was(49.6±3.1)%,the moisture content of feces in the middle dose group of Chinese medicine was(50.4±5.6)%,and the moisture content of feces in the high dose group of Chinese medicine was(58.1±8.2)%.The number of fecal pellets and the moisture content of feces in the low-dose group of traditional Chinese medicine,the middle-dose group of traditional Chinese medicine and the high-dose group of traditional Chinese medicine were higher than those before treatment.The difference in fecal pellets and fecal moisture content between the high-dose Chinese medicine group and the model group was statistically significant(P <0.05),the difference was not statistically significant compared with the normal group(P>0.05).2 Comparison of the discharge time of the first black stool: the discharge time of the first black stool in the normal group was(392.03±5.13)min,and the discharge time of the first black stool in the model group was(620.14±3.39)min,and the first black stool in the low dose group of Chinese medicine was discharged.The time was(605.92±1.38)min.The discharge time of the first black stool in the middle dose group of Chinese medicine was(578.14±7.61)min,and the discharge time of the first black stool in the high dose group of Chinese medicine was(401.57±9.88)min.The discharge time of the first black stool in the low,medium and high dose groups of Chinese medicine was significantly shortened,and the difference was statistically significant compared with the normal group(P<0.01).The first black stool discharge time of the Chinese medicine high-dose group was close to the normal group,and there was no significant difference compared with the normal group(P>0.05).3 Comparison of intestinal propulsion rate: the intestinal propulsion rate of the normal group was(76.03±4.34)%,the intestinal propulsion rate of the model group was(53.71±1.09)%,and the intestinal propulsion rate of the low dose group of Chinese medicine was(62.27±3.12)%.The intestinal propulsion rate of the Chinese medicine middle dose group was(63.02±5.75)%,and the intestinal propulsion rate of the Chinese medicine high dose group was(75.88±1.02)%.After treatment,the intestinal propulsion rate of the low,medium and high dose groups of Chinese medicine was higher than that of the STC model group.The intestinal propulsion rate of the low dose group of traditional Chinese medicine was P>0.05 compared with the model group.The intestinal propulsion rate of the high-dose Chinese medicine group was the closest to the normal group,and the difference was not statistically significant(P>0.05).There was a statistically significant difference between the normal group and the STC model group,P<0.05.4 Comparison of colonic myoelectric activity: The slow wave of the colon in the normal group showed an irregular sinusoidal curve with a frequency of(12.72±1.87)times/min and an amplitude of(0.31±0.02)mv;the STC model group The myoelectricity of colonic rats in 5 rats was significantly slowed down,the amplitude was decreased,the frequency was(8.09±3.84)times/min,and the amplitude was(0.41±0.11)mv;the myoelectricity of colonic cells in 4 rats was significantly accelerated,the amplitude was not equal,the frequency It is(20.67±10.25)times/min and the amplitude is(0.69±0.39)mv;the fast frequency of Chinese medicine low dose group is(25.09±4.44)times/min,the amplitude is(0.45±0.21)mv,and the slow frequency is(6.17±7.09)times/min,the amplitude is(0.22±0.06)mv;the fast frequency of the Chinese medicine middle dose group is(20.68±2.11)times/min,the amplitude is(0.27±0.11)mv,and the slow frequency is(8.23±).1.93)times/min,amplitude is(0.42±0.02)mv;fast frequency of Chinese medicine high dose group is(19.51±6.72)times/min,amplitude is(0.41±0.06)mv,and slow frequency is(12.36±3.61)Times / min,amplitude is(0.37 ± 0.14)mv.Compared with the normal group,the STC model group showed a two-way change in the frequency and frequency of colonic myoelectricity.The difference between the two groups was statistically significant(P<0.05).The high dose of Yiqi Jianpi Tongbian was slowed down.The frequency has a recovery effect and has a mitigating effect on the accelerated frequency.Compared with the model group,P<0.01,the difference is statistically significant.Conclusion: 1 Yiqi Jianpi Tongbian can promote STC intestinal transit movement and defecation;2 The mechanism of Yiqi Jianpi Tongbian Recipe in the treatment of slow transmission constipation may be that it can achieve benign two-way regulation of slow waves of STC colon abnormalities.
