Preliminary Research Of Quality Standard On Homalocladium Platycladum (F. Muell. Ex Hook.) L. H.bailey

Objective:Studyed on breed source,the character,microscopic and TLC identification and determinated the content of moisture,ash,ethanol extract and the content of total flavonoids and quercetin in the quantiative and qualitative study,and researched the fingerprint of Homalocladium platycladum(F.Muell.ex Hook.)L.H.Bailey.,preliminary established quality control method and standard Homalocladium platycladum(F.Muell.ex Hook.)L.H.Bailey.,lay a foundation for its further development and utilization.Methods:The study adopted origin identification,macroscopic identification,microscopic identification,physical and chemical identification to research the pharmacognosical identifying characteristics of Homalocladium platycladum(F.Muell.ex Hook.)L.H.Bailey.,According to the methods stated in fourth volume of Chinese Pharmacopeia(Edition 2015)to research the content limitation control of moisture,ash,and ethanol extract were determined;Used the Orthogonal test to optimize the extraction process of total flavonoids from Homalocladium platycladum(F.Muell.ex Hook.)L.H.Bailey.,by UV-Vis determined the contents of total flavonoids.By HPLC determinating the quercitrin content of Homalocladium platycladum(F.Muell.ex Hook.)L.H.Bailey.and preliminary researched the fingerprint of it.Results:(1)Source identification,character identification,microscopic identification and TLC identification were remarkable,which could be used as reference for the identification of the authenticity of Homalocladiumplatycladum(F.Muell.ex Hook.)L.H.Bailey.(2)The moisture in Homalocladium platycladum(F.Muell.ex Hook.)L.H.Bailey.was between6.39%~10.32%,the total ash was between 4.88%~10.65%,the content of acid insoluble ash was between 0.02%~0.38%,and the content of ethanol extract was between 10.85%~ 28.45%,To its moisture should not exceed 12.0%,the total ash should not exceed 13.0%,acid insoluble ash should not exceed0.5%,the content of ethanol extract should not be less than 9.0%.(3)The optimum extraction process of total flavonoids:Precise weighing of medicinal powder 1g,the extraction solvent was 80% methanol,liquid material ratio1:60,extraction time was 30 min.Rutin showed good linear relationship in the range of 0.0015~0.0368mg/ml,the average recovery was 95.31%,and total flavonoids content was between 13.14~32.41mg/g.regulation of total flavonoids content is not less than 0.008%(mg/g).(4)the quercetin showed good linear relationship concentration in 0.023~0.460mg/ml,the average recovery rate was 100.86%,The quercitrin content of Homalocladium platycladum(F.Muell.ex Hook.)L.H.Bailey.was between0.05~0.50%.regulation of quercitrin content is not less than0.004%.(5)Established Homalocladium platycladum(F.Muell.ex Hook.)L.H.Bailey.HPLC fingerprint,screened 11 common characteristic peaks,Dentified 10 as quercitrin,and taked it as the reference peak detection,the 20 batch ofmedicinal materials and the fingerprint similarity between 0.924~0.992.Conclusions:The evaluation methods were simple,had good repeatability,obvious characteristic,which provide reference for the revision of the quality standard of Homalocladium platycladum(F.Muell.ex Hook.)L.H.Bailey.,and laied the foundation for controling safe,effective and quality of Homalocladium platycladum(F.Muell.ex Hook.)L.H.Bailey.on the clinicalapplication.
