Chicory is the dry aboveground part or root of Cichorium glandulosum Boiss.et Huet or Cichorium intybus L.It has the effects of clearing the liver and gallbladder,strengthening the stomach and reducing swelling.Due to the imperfect quality standards of chicory,it is difficult to control the quality of the herbs at present.The quality standard of Cichorium intybus L was listed as an issue raised by the standard of Chinese Pharmacopoeia in 2020.Therefore,it is necessary to improve and complete the quality standards of Cichorium intybus L.The research contents of this subject are as follows:1)A method for microscopic characterization of chicory powder was established and described.According to the method of pharmacopoeia,the purchased chicory medicinal materials were characterized for trait identification and thin layer identification(TLC).The impurities,moisture,ash,and extracts of chicory were examined,the limits of which were also established.2)Through studying ten batches of chicory roots from different areas,the characteristic chromatographic methods of chicory roots were established by high performance liquid chromatography.The chromatographic column was AlltimaTMM C184.6×250 mm column;The column temperature was 30°C;The detection wavelength was at 254 nm;The injection volume was 10μL;The flow rate was 0.9 mL/min;The acetonitrile was mobile phase A,and the 0.05%phosphoric acid aqueous solution was mobile phase B;Gradient elution.Methodological validation of the feature map method was carried out.Experiments showed that the method had strong specificity and good repeatability.The quantity and species of chemical constituents could be reflected by the characteristic Atlas of chicory roots,which made the comprehensive evaluation of the internal quality of chicory roots,and provided a basis for the comprehensive control of chicory quality.3)Establish a method for determining the content of chicory root and validate the method.The contents of ten batches of chicory root were also determined.The chromatographic column was AlltimaTM C18 4.6×250 mm column;The column temperature was 30°C;The detection wavelength was at 328 nm;The injection volume was 10μL;The flow rate was 1 mL/min;The mobile phase was methanol and 0.1%phosphoric acid solution(25:75).The limits for determination of chicory root content were established.According to the calculation of dried products,the contents of chlorogenic acid should not be less than 0.03%.The method had strong specificity,good repeatability and stability within 24 h.The determination of chicory root content effectively controled the quality of chicory root,which provided a basis for the quality standard of chicory root.4)The polysaccharides of Cichorium intybus L were extracted by ultrasound.The content of polysaccharide in Cichorium intybus L was determined by phenol-sulfuric acid colorimetric method,and the experiment of antioxidant activity in vitro was carried out.The results showed that the content of polysaccharides in ten batches of chicory root was2.66 mg/g5.94 mg/g.The contents of polysaccharides in the aerial parts of two batches of chicory were 4.76 mg/g and 5.24 mg/g.The results showed that polysaccharides in chicory had antioxidant activity in vitro.In this study,the quality standard of chicory was studied comprehensively,which provided a basis for the quality standard of Cichorium intybus L. |
