A Kidney-tonifying Herbal Fufang Treatment Impact Bone Mineral Density Of Senile Osteoporotic Mice By GH/IGF-1 Axis

Objective:(1)to detect the kidney-tonifying herbal Fufang for rapid aging male mice's serum concentration of GH and IGF-1 and local bone tissue the expression of GH mRNA and IGF-1 mRNA.(2)analysis the correlation of bone mineral density respectivelyand the serum concentration of IGF-1 and GH and the expression level of GH mRNA and IGF-1 mRNA in local bone tissue.Methods:selected 40 only 3 months rapidly aging male osteoporosis model SAMP6 mice(P6),and 10 normal aging SAMR1 homologous comparison male mice as a research object.P6 is divided into four groups,each group of 10,only with bone kidney-tonifying herbal Fufang,rhGH subcutaneous injection,kidney-tonifying herbal Fufang+ rhGH subcutaneous injection,the amount of normal saline lavage,SAMR1 irrigation amount of saline.Experimental animals in two batch of materials,each batch of random take half,respectively was conducted in the experiment after 3 months and 6 months,detect the bone density,to eyeball take blood,and to detect serum GH,IGF-1 and bone turnover markers by ELISA method and detect the expression level of GH mRNA and IGF-1 mRNA in bilateral tibial by qPCR,statistical data analysis in the end.Results:(1)of mice P6 empty,body hair is thin and yellow began to emerge,depression,burnout lethargy,bite,lavage owing to cooperate,shit.RhGH group hair hair is sparse,owe luster,spirit is a bit poor,bite,lavage owing to cooperate,shit.Kidney and kidney + rhGH group,R1 group,responsive,activity,behavior dial,lavage,shit.Each weight during the experiment were increased with the extension of feeding time.(2)3 months after the intervention found that serum concentration of CTX-1,P1NP and BGP in rhGH group is higher than empty P6;Kidney group serum concentration of CTX-1 drop when compare with empty P6 group,serum concentration of P1NP and BGP are up;Kidney+rhGH intervention group,serum concentration CTX-1,P1NP and BGP are rising in the P6 empty group.Six months after the intervention:the serum concentration of CTX-1 drop in rhGH group when compare with P6 empty group,serum concentration of P1NP and BGP increased compare with P6 empty group;Kidney group serum concentration CTX-1 drop compare with P6 empty group,serum concentration of P1NP and BGP up;Kidney + rhGH group serum concentration drops compare with the P6 empty group,the concentration of CTX-1,P1NP and BGP are rising in the P6 empty group.(3)after 3 months intervention,we found that serum concentration of GH and IGF-1 in the drug intervention group with different degree rising,serum GH concentration with rhGH group was obviously higher than that of kidney and kidney+rhGH group;Three groups of serum IGF-1 there was no statistically significant difference.Six months after intervention:rhGH group,kidney,kidney + rhGH group compared with the blank group,the serum GH,concentration of IGF-1 has increased,with significant difference statistically significant,but the three groups of drug intervention group there was no statistically significant difference comparing the serum concentration of GH,kidney+GH and kidney group serum concentration of IGF-1.(4)rhGH group,bone tissue in GH mRNA and IGF-1 mRNA expression level increased when compare with the P6 empty group,,the expression level and kidney group were lower than rhGH group,kidney+rhGH group compared with GH and kidney group expression level is high.(5)Spearman correlation analysis results show that the concentration of serum GH,IGF-1 respectively with bilateral tibial GH mRNA,IGF-1mRNA expression levels were positively-correlated;Serum concentration of GH and IGF-1 serum concentration was significantly positive correlation with Bilateral femoral GH gene expression level and IGF-1 are positively.(6)After 3 months intervention,rhGH group compared with P6 blank group,rhGH group of whole body,femur and spinal bone density difference compared with the P6 blank group was not statistical significance;Kidney set of whole body and femoral bone mineral density and higher than the P6 empty and rhGH group,difference has significant meaning;Kidney+rhGH1 s BMD is highger than P6 empty group,the differences are statistically significant meaning;when compare the Kidney+rhGH group to rhGH and kidney group,two groups of bone mineral density difference is no statistical significance.Six months after intervention:rhGH set of whole body and femoral bone mineral density is high when compare to the P6 blank group,but with significant difference statistically significant;Kidney group of whole body,femur,spine,and pelvis bone mineral density is higher than the P6 blank group with differences statistically significant;Kidney group body,spine and pelvis bone density than rhGH group is high,no statistical significance difference;Kidney+rhGH group of whole body,femur,spine,and pelvis bone density are higher than the P6 blank group,the differences are statistically significant meaning;Kidney+rhGH pelvis bone density than rhGH group is high,the difference is statistically significant,the difference compared with kidney and R1 groups has no statistical significance.(7)6 months after treatment,Spearman correlation analysis results show that the concentration of serum GH and the whole body,femur,P6 mice is closely relative to the spine and pelvis bone mineral density,the concentration of serum IGF-1 and P6 mice body,bone mineral density is related to spine and pelvis.Conclusion:(1)bone mineral density,bone turnover index measurement results show that the P6 conforms to the characteristics of senile osteoporosis in mice,as the growth of the age and P6 mice serum GH,concentration of IGF-1 decline,in line with the requirements of our research.(2)recombinant human growth hormone replacement therapy intervention for 3 months before the P6 mice bone mineral density may be the same or even lower,may be due to GH promote bone formation and increase osteoclast activity,prolong intervention to bone mineral density increase was observed after 6 months,showed that after 6 months intervention rhGH osteogenesis effect is greater than the broken bone.(3)kidney strong bone grain intervention 3 and 6 months can be observed that rising of the concentrations of serum GH,IGF-1,six months after intervention serum GH,concentration of IGF-1 and GH intervention group had no significant difference,kidney strong bone granules could instead of recombinant human growth hormone in the treatment of decreased of GH and IGF-1 with increasing age.(4)recombinant human growth hormone,although joint kidney strong bone grain intervention in the intervention six months when rising serum IGF-1 respectively than single intervention to rise faster,but bone mineral density increase is not obvious difference was observed,may be combined treatment not bring greater benefits.(4)6 months after the intervention,the serum GH,concentration of IGF-1,bone tissue GH mRNA,IGF-1 mRNA expression level and each part of the whole body bone mineral density were positively correlated.(4)kidney strong bone particles in rapid aging mice bone mineral density and increased bone tissue of GH and IGF-1 expression at the same time can improve the quality of the survival of mice.
