ObjectiveFructus Amomi,as a commonly used traditional Chinese medicine,the market demand is big.In view of the current medicine market in Fructus Amomi's quality is muddledness,dragons and fishes jumbled together and shoddy phenomenon are very serious,it has a great impact on the production,planting and market of the high qualityand and real Fructus Amomi.Based on the theory of "Bian-zhuang-lun-zhi" this study will be using the intelligent sensory technology combine with modern chromatographic analysis technology,to achieve digitization of the fuzzy information of digital gas and the flavour of Fructus Amomi,comprehensive quality evaluation of Fructus Amomi from appearance,chemical fingerprint,odor characteristics,application of numerical techniques of mass spectrometry of multi component qualitative analysis and pharmacodynamic activity.At the same time,the rapid identification of electronic nose and tongue establishment method of Fructus Amomi,provide the basis for further improve the quality evaluation standard of Fructus Amomi,to promote resource development and protection of the varieties of Fructus Amomi.Methods1.Fructus Amomi's market,origin investigation and character identification.we collected a total of 22 batch of Fructus Amomi from domestic and imports via the origin of acquisition and marmet purchases;we take the traditional empirical identification methods,invite experts to conduct charater identification and grading of Fructus Amomi that we collected.2.The quality evaluation of Fructus amomi based on chemical components.Respectively on the main chemical components of 22 batches of samples of volatile oil,bomyl acetate and quercitrin were determined;The establishment of the HPTLC fingerprint of Fructus Amomi,to evaluate the quality of the collected samples of Fructus Amomi.3.A comparative stydy based on Fructus Amomi's raw powder "gas".The aroma chatacteristics of Fructus Aromi from different origin were investig ated using electronic nose and automatic headspace-gas chromatography-mass sp ectrometry(HS-GC/MS)from two aspects of the overall aroma profile and spe cific aroma composition,a bioplot was used to reveal the correlation among the sa mple,E-nose gas sensors,and the chemical volatile compounds,to build objective,rapid and digital method of Fructus Amomi 's quality evaluation about electronic nos e technology.4.A comparative study based on Fructus Amomi aqueous extract's "flavour".We adopts the methods of electronic gongue combined with principal componen t analysis to analyse the electrical signal data collection.Based on HPLC-Quard TOF MS/MS combinedwith multivariate statistical analysis technology for different r egions Fructus Amomi aqueous extract to compare the chemical composition analysi s,find out the differentorigin of Fructus Amomi aqueous extract differences in che mical composition,try toclarify the material base of Fructus Amomi aqueous extract's "flavor".5?A comparative study of the Fructus Amomi in yunnan and in Burma aqueou s extract which effect of human gastric cancer cells SGC-7901 apoptosis.Use four methyl thiazolyl tetrazolium(MTT)method to compare the inhibiton effect of Yunnan and Burma Fructus Amomi aqueous extract on the proliferation of gastric cancer SGC-7901.Hoechst33342 staining was used to observe the apoptosis of SGC-7901 cells.Use the flow cytometry to detect the impact of cell apoptosis and cell change cycle of SGC-77901 induced by Fructus Amomi aqueous extract.Results1.we collected a total of 22 batch of Fructus Amomi herbs,of which 7 batches of Yunnan,Guangdong 4 batches,the 6 batches of Burma,Vietnam 3 batches,Laous 2 batches.The survey results show that the yield and planting area of Fructus Amomi in Guangdong and Yunnan decreased in recent years,lead to sustained high prices,but the Chinese market sales to Southeast Asia imported Fuctus Amomi,has a low prise.The properties identification method evaluate the quality of 22 batches of Fructus Amomi preliminarily,which is diviede into three grades:first grade for the fruit ovoid,surface soft burrs invertebrates,seeds oil moisten soft glue,gas concentration,taste cool feeling obviously,have hot feeling.Amomum villosum Lour.from Guangdong and Yunnan are the first grade;Fruit ovoid plumpness is not enough,the second grade for less surface soft burrs,seed oil moisten degrees lower,aroma,taste sheen cool feeling lighter.The two batches of Laos are second grade;Third grade for oblong fruit peel hard,burr short and little,seed dry,no gas,taste not cool feeling.The Fructus amomi from Burma and Vietnam are third grade.2.Content determination results of the primary efficacy component:the total content of volatile oil results in Amomum villosum Lour.from Yunnan and Guangdong were higher than import Fructus Amomi volatile oil content,including Jinghuakeng to a maximum of 7.7±0.2%of the original region,Fructus Amomi from Burma have 5 batch of unqualified;Bornyl acetate content determination results showed 6 batch of Fructus Amomi in Burma are unqualified,Amomum villosum Lour.from Yunnan and Guangdong bornyl acetate content higher than imported Fructus Amomi;Quercetin content determination results between different batches Fructus Amomi medicinal materials without a certain regularity,in the range between 0.0098 to 0.0682 mg/g.The fingerprint of Fructus Amomi HPTLC established can visually see the varying degree differences in the relative content of each batch of Fructus Amomi maker components,similarity evaluation,clustering analysis and principal component analysis results will be divided into 3 classes of Fructus Amomi.,the overall chemical compositon of Amomum villosum Lour from Yunnan and Guangdong is closed,villosum and amomum villosum overall chemical composition,the differences between Burma Fructus Amomi and Amomum villosum Lour about overall chemical compositon is the largest.3.The E-nose showed a good performance in grouping with a principle component analysis explaining 83.9%of variance.The homebred Amomum villosum is classified as a class,and the import of Fructus Amomi is classified as a class.A total of 70 aroma components were identified by HS-GC/MS,with terpenes,esters,alcohols,ketone and alkane.It was observed that the components and the number of terpenes,esters and alcohols were different in the different origin of Fructus Amomi.The results of pattern recognition confirmed that HS-GC/MS processing results were similar to that of E-nose.A bioplot of PLS was introduced to display the high correlation between sensors S14/S8/S6/S3 and terpenes,esters,alcohols indicated that special sensors were highly related to some aroma constituents.4.The electronic tongue PCA analysis results can distinguish adulterants shellfl ower from Fructus Amomi,but with different origin and different varieties of Fruct us Amomi to distinguish the effect is not ideal;The difference research based on hi gh performance liquid chromatography-tandem level 4 pole flight time high resolu tion mass spectrometry in the different region Fructus Amomi aqueous extract,Fruct us Amomi from different production areas can be correctly distinguish,to some exte nt,the classification results reflects differences between Fructus Amomi in the regio n,received 26 kinds of differences in composition,infer that the differences of 11 kinds of compounds,including the phyllopod shirt coumarin,genistein glycosides,(1 r,2s,5s)-6-oxa-3-azabicyclo[3.1.0]hexane-2-carboxylic acid,1,6-beta-D-dehydration of glucose,vanillic acid-l-beta-D glucose ester glycosides,gentian acid,p-hydroxy benzoic acid,epicatechin,isoquercetin,quercetin,3,5-Dihydroxy-7-(4'-hydroxy-3'-meth oxyphenyl)-1-(3',4'-dihydroxyphenyl)heptane.5?Fructus Amomi from Yunnan and Burma,their aqueous extract can inhibit the proliferation of tumor cells SGC-7901,concentration dependence,determined by MTT method,Hoechst33342 staining and flow cytometry results are shown in Yunnan Fructus Amomi aqueous extract that apoptosis rateof SGC-7901 tumor cell proliferation inhibition higher than Burma Fructus Amomi water extract,The cell cycle experiment results indicate Yunnan Fructus Amomi aqueous extract to block SGC-7901 cells in G2/M phase,while the Burma Fructus Amomi aqueous extract block in S phase.ConclusionThis study based on the theory of "Bian-zhuang-lun-zhi",through the appearanc e,gas and flavor and combined intelligent sensory technology with the modern chro matographic analysis to analyzed the overall quality evaluation of Fructus Amomi.A nd the contrast experiment about pesticide effect was took on optimized Yunnan Fru ctus Amomi and poor quality Burma Fructus Amomi.Research results show that the quality of Fuctus Amomi fruit appearance was oval,top fruit connecting surface is not obvious in brown,triquetrous,round with soft spines,fragrant smell,taste swe et,spicy,sour,bitter and salty bittersweet,with pungent and cool feeling bitter.The electronic nose used in this study feature extraction of Fructus Amomi aromo,esta blish the relationship between the coupling between quality and olfactory informatio n,realize the digital information so that the aroma of Fructus Amomi,the traditiona l morphological identification experience scientific and objective.The results show that no significant differences between Yunnan and Guangdon g geohebs in Amomum villosum Lour.about sensory properties,the overall chemical composition,quality is very close,and the import price of Fructus Amomi although cheap but in terms of appearance,chemical composition,and the overall efficacy was far from Amomum villosum Lour..In recent years,Yunnan province in the Amo mum villosum Lour.cultivation management increasingly standardized,geographical en vironment and the advantage of planting,the Yunnan area of Amomum villosum Lou r.yield,moderate price,this paper provides a theoretical basis for the promotion of r esource development in Yunnan province of Fructus Amomi.
