The Curative Effect Experiment Study Of Fructus Amomi Rotundus And Combined With 5-Fu To Treat The Gastric Cancer

[Objectives]Cancer damage human health seriously only next to heart trouble,and the incidence of it is increasing every year.The tumor researchers are considering the new ideas,new ways and new methods to cure tumor over a long time.The traditional Chinese medicine is the valued fortune of our country.There are rich natural medicine materials in our country,and tumor researchers attach more importance to the effect of these medicine materials.Using the traditional Chinese medicine to cure tumor is becoming one of the most important ways.The purpose of this study is to identify the influence of fructus amomi rotundus (FAR)combine with 5-FU on growth and metastasis of gastric cancer in an orthotopic model of nude mice,and go a step further to lay a foundation of studying clinical usage that FAR resist tumor .[Methods]The experiment bases on 24 nude mice model of orthotopic transplantation of human gastric cancer.They are divided randomly into four groups(six in each):control group;5-fluorouracil group(5-FU group);FAR group,Combination group:5-FU+FAR group.The control group pours 0.2ml PBS into the stomach,qd×4wk;and injects 0.2ml PBS into the abdomen,q2d×7day.The 5-FU group pours 0.2ml PBS into the stomach,qd×4wk;and injects 12.5mg 5-FU into the abdomen,q2d×7day.The FAR group pours FAR (1g/kg·d)into the stomach, qd×4wk; and injects 0.2ml PBS into the abdomen,q2d×7day.The combination group pours FAR (1g/kg·d)into the stomach, qd×4wk; and injects 12.5mg 5-FU into the abdomen,q2d×7day.Among the experiment,observe animal action and record animal weight every day.At the end of four weeks of the experiment,volume,weight,pathological changes of tumor tissues,local invasiveness and metastasis in the local lymph node,liver and spleen are compared among different groups.[Results]1.Sucessfully build a stable orthotopic implantion model of gastric cancer in nude mice.2.Tumor volume is(1236.31±123.77)mm3 in Control group;(509.95±56.07)mm3 in 5-FU group(P<0.05); (739.72±75.86)mm3 in FAR group(P<0.05);(235.88±21.37)mm3 in Combination group(P<0.05)。3.The inhibition rate of 5-FU group,FAR group,Combination group is 55.99%,42.81%,67.36% respectively.4.The weight of agent therapy groups is obvious lower than Control group.5.In Combination group,the incidence of local invasiveness and metastasis in the local lymph node,peritoneum and liver are significantly lower than those of Control group and single-agent therapy groups.[Conclusions]1. 1g/(kg﹒d)FAR which pours into the stomach can restrain the growth of BGC-823 tumor cell.2. Synergistic effect is achieved by combination of FAR with 5-FU,it shows more remarkable inhibition on the tumor growth and metastasis of orthotopic xenograft of BGC-823 cells in nude mice.
