Objective Adult male Sprange-Dawley(SD)rats were used to establish a model of trigeminal neuralgia(TN)through the operation of chronic constriction injury of the infraorbital nerve(ION-CCI).The behavioral experiments were tested to discriminate whether the model was set up successfully.Western blot test was used to detect the IL-17 and P-P38 protein expression changes and RT-PCR test was used to detect the m RNA expression of IL-17 A,CXCL1 and CXCL2 in trigeminal ganglion(TG)in order to discuss the mechanism of IL-17 and P38 signal path in the ION-CCI TN model rats.Methods Male SD rats were randomly assigned into two groups,ION-CCI group: The chronic constriction injury of the infraorbital through the lower edge of cheekbone was operated to establish the TN model,and sham group: Infraorbital nerves were only exposed but not ligated.Before operation,and 1 w,2 w and 3 w after the operation,the mechanical withdrawal thresholds of vibrissa pad in two groups of rats were measured respectively.The recorded results were compared to determine whether established the TN model successfully.Total protein of IL-17 and P-P38 in TG were extracted and detected by Western blot test to half quantitatively analyze the grey value.Total m RNAs of IL-17 A,CXCL1 and CXCL2 in TG were extracted and detected by real-time fluorescent quantitative PCR test.All data were analyzed statistically by SPSS17.0software.Results Compared with sham group,the mechanical thresholds of ION-CCI group were obviously decreased,and at 2 week after operation,the mechanical thresholds of ION-CCI group were the lowest(sham group: 14.11 ± 0.81 g vs ION-CCI group: 4.45 ±0.92 g,n=5,P < 0.01).The IL-17 and P-P38 protein in TG of ION-CCI group over-expressed distinctly after operation,and they were the highest at 2 week after operation,which significant difference compared with that before operation(n = 3,P <0.05).Compared with sham group,the IL-17 A and related chemokines(CXCL1,CXCL2)m RNA in TG of ION-CCI group over-expressed distinctly also at 2 week after operation.The m RNA expressions of IL-17 A of ION-CCI group were significant different compared with that of sham group(n = 3,P < 0.05).Conclusions The IL-17 has an important role in the developing of TN.The IL-17 may lead to the TN through the P38 signaling pathway.
