| Objective: To evaluate on authenticity the genuine and inferior Merremia hederacea(Burm.f.)Hall.f.and establish initially quality standard draft of Merremia hederacea(Burm.f.)Hall.f.by deeply and systematically studying.Methods:(1)Pharmacognostic qualitative study of Pharmacognostic of Merremia hederacea(Burm.f.)Hall.f.was systematically carried out by origin character,microscopic and physical and chemical identification methods.(2)According the methods stated in the fourth volume of Chinese Pharmacopeia(2015 edition)to research its examination of limited,including moisture,total ash,acid-insoluble ash and extract and so on.(3)To study the chemical components of Merremia hederacea(Burm.f.)Hall.f.from three origin by the methods of systematic pretest.(4)The total flavonoid was determined by applying UV-vis spectrophotometry method,and using rutin as comparative samples.(5)With chlorogenic acid as the reference substance,determining the content in Merremia hederacea(Burm.f.)Hall.f.of different habitats by HPLC and confirming its limit of chlorogenic acid.(6)At the same time,establish the bioassay method based on antibiotic effect.Results:(1)The pharmacognosical characteristics of origin identification,macroscopic identification,microscopic identification,physicochemical identification all are remarkable,so the pharmacognosical characteristics can be used for the identification of Merremia hederacea(Burm.f.)Hall.f..(2)Results showed that the moisture content of Merremia hederacea(Burm.f.)Hall.f.fluctuated from 6.26% to 9.96%,therefore moisture content not more than 11.00%.The total ash and acid-insoluble ash fluctuated from 6.31% to 11.55% and 0.20% to 1.60%,therefore the total ash and acid-insoluble ash not higher than 13.00% and 1.70%.Based on the extractions fluctuating from 4.69% to 9.22%,the extractions not be less than4.00% could be tentatively given.(3)The Results showed that Merremia hederacea(Burm.f.)Hall.f.contains flavonoids,polysaccharides,glycosides,alkaloids,anthraquinones,proteins,amino acids,organic acids,tannins,phenols and other chemical constituents.(4)The contents of total flavonoids fluctuated from 5.10% to 14.32%.Based on the amounts,the amounts of total flavonoids not less than 4.00% could be tentatively given.(5)The contents of chlorogenic acid fluctuated from 0.22% to 0.64%.Based on the most amounts,the amounts of chlorogenic acid not less than 0.20% could be tentatively given.(6)There was a linear relationship between dose and effect in special concentration range with good crrelation,and the bioassay potent was initially defined as ?0.8000U/ML.Conclusion: The research results provided theoretical evidence and data for accurately identifying and establishing the quality standard of Merremia hederacea(Burm.f.)Hall.f.,and for utilizing Merremia hederacea(Burm.f.)Hall.f.. |
