Abstract/Summary:
Polysaccharides from Inonotus obliquus have no toxic effect with cutting downblood sugar, anti-tumor,cutting down blood fat,anti-oxidation. At present,study onInonotus obliquus and its polysaccharides in China are unsubstantial. There is littlereport on isolation,purification,structure composition and system study of cutting downblood sugar of polysaccharides from Inonotus obliquus.In this paper,based onextraction and preliminary purification of polysaccharides from Inonotus obliquus,their monosaccharide composition and cutting down blood sugar effect were furtherinvestigated.The first chapter firstly summarized the morphology,growthenvironment,chemical composition,pharmacological effects of Inonotus obliquus.Thenintroduced the present study of extraction,deproteintion,bleach and the mechanism ofhypoglycemic of Polysaccharides from Inonotus obliquus. Thus determined thepurpose and significance of the thesis topics.In the second chapter, the single factor experiment was adopt to analysis fivefactors such as cellulose enzyme dosage, reaction temperature, solvent times theamount of reaction time, pH, impacting polysaccharide extraction, and the orthogonalexperiment was adopted to optimize the conditions affecting cellulase-assistedextraction of polysaccharides. The experiment ultimately improved the extraction rateof polysaccharides from Inonotus obliquus, got the best possible extraction solution.A variety of different deproteintion,bleaching methods were studied in the third chapter,by compared their rates of protein removal, pigment removal rate, polysaccharide retention rate,and ultimately determined the best deproteinized andbleaching solutions,got the high purity refined polysaccharides from Inonotus obliquus,and can be used providing the basis for analysis the monosaccharide hydrolyzed from Inonotus. The conditions of the optimal hydrolysis,the optimal derived,and and temperature-programmed gas chromatography analysis were obtained by studying in thefourth chapter.Then determined the monosaccharide composition and proportionin the Polysaccharides from Inonotus obliquus.In chapter v, we discussed different dose groups of Inonotus obliquus polysaccharides which match the corresponding glucose consumption and cell proliferation of HepG2cells through the cell in vitro experiment. It was proved thatInonotus obliquus polysaccharides had good hypoglycemic effects in vitro. Also,we got a preliminary conclusion that its hypoglycemic effects may be mediated by improving glucose metabolism of peripheral tissue and the sensitivity of liver cells to insulin.
