Objective In this study,we had discussed the mechanism between obese with benign prostatic hyperplasia on a sustainable moderate-intensity exercise and diet,and emphasizedadipocytokines and sex hormones as a key mediator under the background of benign prostate hyperplasia induced by obese mice.Therefore we can afford new evidences to prevent and treat benign prostatic hyperplasia induced by obese.Methods80 male C57BL/6J mice were assigned into 2 groups as a 16 mice for normal diet group,and 64 mice for high-fat diet group.After 8 weeks,there were 38 mice have met the weight standards.8 mice were dissected and cutted tissues in each group as C group(Control group,C)and Ob group(obese group,Ob).Analysis of glucose and lipid metabolism and relevant indicators of benign prostatic hyperplasia.Then we used sustainable moderate exercise and diet intervention.Those obese mice were divided into Di group(Diet group,Di)and Ex+Di group(Exercise +Diet group,Ex+Di),Co group(Contral group,Co),8 mice in each group were fed with normal diet.Exercise mathods: used a sustainable moderate treadmill exercise,12m/min,60 min,6d/week,6 weeks.The changes of glucose and lipid level and sex hormones level was detected by enzyme liked immunosorbent assay.The changes of prostate tissueswas detected by HE statining.The level of adipocytokines was detected by suspension biochip technology.Expression of Er?and Er?was detected by immunohistochemistry.Statistical methods includes t test,one-way ANOVA and pearson correlation analysis.The experimental results was expressed by (?)±s,r expressed the correlation,the significance of difference was expressed by P<0.05.Results1.There were 38 obese mice had reached obese standards,and the obese rate was60%.The mean level of weight,internal fat in Ob group was higher than C group(P<0.05).and glucose-lipid had happened to be metabolism disorder.All the obese-related indicators indificated the access of the model in Ob group.2.The mean level of prostate weight,prostate index,prostate volume was significantly higher than C group(P<0.05).The mean level of prostate luminal area and prostate epithelial height in Ob group was significantly larger than C group(P<0.05).Prostate pathological sections showed evident pathological changes of prostate hyperplasia in obese mice.All indicators showed that obese mice happened to benign prostate hyperplasia.3.The mean level of E2 and T in the Ob group was higher than the C group(P>0.05).but the rate of E2/T in obese group was higher than C group(P<0.05).The analysis of pearson correlation showed that there was no correlation between glycolipide,internal fat with prostate volume,prostate index in Ob group(P>0.05).4.Exercise intervetion:The level of mice weight,internal fat in Ex+Di group were significantly decreased(P<0.05),The level of glucose-lipid in three group was no significant difference(P>0.05).Excese had improved glucose-lipid.5.The level of prostate weight,prostate index,prostate volume in Ex+Di group was significantly lower than the level of Di group(P<0.05).and compared to Co group,there was no significant difference(P>0.05).Prostate pathological sections showed evident pathological changes of prostate hyperplasia in Di group.But exercise and diet improved the hyperplasia of prostate in Ex+Di group.6.The analysis of pearson correlation showed that there was a positive correlation between visfatin with prostate volume,but there were no correlation in other indicators.The level of visfatin,resistin in Ex+Di group was significantly lower than Di group(P<0.05),and compared to Co group,there was no significant difference(P>0.05).The level of glucagon was no significantdifference in each group(P>0.05).7.There were no significannt difference in E2,T and the rate of E2/T in each group(P>0.05),but they had a decrease trend.The level of AOD of ER? in Ex+Di group was significantly lower than Di group(P<0.05),and there was no difference between Ex+Di group with Co group(P>0.05).The level of AOD of ER? in Ex+Digroup was significantly higher than Di group(P<0.05),and there was no difference between Ex+Di group with Co group(P>0.05).Exercise and diet had up regulated the level ER?,and reduced the level of ER?.Conclusion The obese mice happened to benign prostate hyperplasia which were fed by 60%high fat diet,but the weight were reduced in obese mice by exercise and diet.Exercise and dietimproved the minienvironment of benign prostate hyperplasia by regulating adipocytokines and sex hormones.
