ObjectiveA large number of epidemiological and disease health data show that increasing the intake of whole grain food can effectively reduce the risk of many chronic diseases.At home,most studies on whole grains concentrate on the analysis of certain functional ingredients.Experimental and epidemiological studies on rye whole wheat flour did not be found and all rye whole wheat flour quality on the market did not meet the corresponding requirements.Besides,the nutritional value of rye whole wheat flour is higher.Based on this,this paper takes the rye whole wheat flour as a whole test,performing animal experiments.Discuss the regulatory effects of two different processing techniques rye whole wheat flour on lipid metabolism disorder rats and hyperglycemia mice,besides,analyzing its possible mechanism MethodsHypoglycemic test:Two hundred adult male Kunming mice were purchased.After two days adaptive feeding,the mice were injected fresh alloxan(130 mg/kg BW).The blood sugar level higher than 10mmol/L and less than 25mmol/L was hyperglycemia model animals.The hyperglycemia model animals were randomly divided into seven groups:one control group and six dose groups,including three common rye whole wheat flour groups that were low dose group(group A),middle dose group(group B)and high dose group(group C)and three rye flour of complete method which were low dose group(group D),middle dose group(group E)and high dose group(group F).Continuous feeding six weeks,records the general situation of animals.After the end of the experiment,fasting for 3-5 hours,takes blood in the tail,measuring fasting blood glucose(FBG)and blood glucose curve area.Using curved tweezers extracts the eyeball,collecting blood,measuring fructosamine(FMN),liver glycogen(HG),urea nitrogen(UN),creatinine(Cre),insulin(INS)and c-peptide(C-P).Blood lipid test:Eighty-five rats were randomly divided into two groups according to body weight.Ten rats were fed with normal diet as blank control group,and seventy-five rats were fed with high fat diet as model control group.After modeling,the model animals were randomly divided into seven groups according to TC level and body weight,that were,model control group and six dose groups,including three common rye whole wheat flour groups that were low dose group(group A),middle dose group(group B)and high dose group(group C)and three rye flour of complete method which were low dose group(group D),middle dose group(group E)and high dose group(group F).Each group is ten rats and given the corresponding test substance.After the end of the experiment,the rats were killed and collected blood,measuring serum or plasma total cholesterol(TC),triglyceride(TG),high-density lipoprotein(HDL),low density lipoprotein(LDL),nitric oxide(NO),thromboxaneB2(TXB2),6-ketone-alpha prostaglandin F1 alpha(6-keto-PGFl?),lipoprotein lipase(LPL),hepatic lipase(HL).The same part of the liver of each group was cut and placed in 10%formaldehyde solution,using paraffin to make the pathological section.ResultsHypoglycemic test:1)Diabetic mice obviously shows the disease characteristics.Model control group and each dose group mice body weight was significantly lower than blank control group(P<0.05).Each dose group mice weight is lower than the model control group and there was no statistically significant difference(P>0.05).2)In addition to the group A,the difference was statistically significant after the intervention of FBG,the group B,C,D,E(P<0.05),group F(P<0.01).3).After the intervention,every test index showed obvious changes compared with model control group,Among them,group B,C,F area under the curve of blood sugar decreased(P<0.05),group C,D,E,F serum FMN level decreased significantly(P<0.01 or P<0.05),group A,B,C,D,E,F serum HG decreased(P<0.01 or P<0.05),B,C,E,F group serum INS increased(P<0.05),group A,B,C,D,E,F serum C-P is no statistical difference(P>0.05),but tend to rise,group B,C,E,F plasma UN decreased(P<0.01 or P<0.05),group B,F serum Cre decreased(P<0.05).There was no significant difference in each test results of each dose group between the rye wheat flour of complete method and ordinary rye whole wheat flour.Blood lipid test:2)The rats weight in model control group and all dose groups were obviously lower blank control group(P<0.05).All dose groups rats weight were lower than the model control group(P>0.05).2)Compared with model control group,each dose group showed certain dose-effect.Among them,C,F group Serum TC decreased(P<0.05),group A,B,C,D,E,F serum TG decreased(P<0.05),group C,E,F serum HDL elevated(P<0.05),all dose groups serum LDL is no statistical difference,but a downward trend,group F serum NO level(P<0.05),group A,C,E,F serum TXB2 decreased(P>0.05),each dose group serum 6-keto-PGF1 alpha changes is no statistical difference,but a downward trend,group D,E,F of liver tissue LPL content increased(P<0.05),group E,F of liver tissue HL content increased(P<0.05).3)Pathological test results show that each dose group compared with model control group,liver cell fat change is degeneration and fat cavitation is less,the nucleus squeezed shift phenomenon gradually disappear.After the intervention,the dose-effect relationship is obvious.There was no significant difference in each test results of each dose group between the rye wheat flour of complete method and ordinary rye whole wheat flour.ConclusionsTwo different processing techniques rye whole wheat flour can effectively reduce the experimental diabetic mice fasting glucose and fructosamine levels,increase the tolerance to glucose,and play a good protective effect in diabetic renal injury caused by alloxan.Its mechanism may be to promote the secretion of insulin,synthesis of liver glycogen and enhancement of glomerular filtration function to improve the hyperglycemia symptoms in miceTwo different processing techniques rye whole wheat flour can obviously decrease the serum TC and TG and elevate serum HDL and there is a tendency to decrease serum LDL.Besides,it can effectively regulate the lipase metabolism,enhance vascular endothelial active substances and prevent the further injury of liver tissue caused by abnormal lipid metabolism.It has the function of improving lipid metabolism disorder.Rye wheat flour of complete method relative to ordinary rye whole wheat flour in improving disorder of lipid metabolism and decreasing blood sugar were no statistical differences,but its experimental results measured values were better than that of ordinary rye whole wheat flour,and because of its good processing technology----improve the quality of the whole wheat flour,prolong the shelf life and lower cost.,Rye wheat flour of complete method has certain biological value and market value.
