Study On The Chemical Constituents Of The Two Kinds Of Chinese Medicine In Health Food Raw Materials

This thesis engaged in the research of chemical constituents of two traditional Chinese medicine in health food raw materials—Astragalus membranaceus(Fisch.)Bge.Var.Mongholicus(Bge.)Hisao and Gardenia jasminoides Ellis,as well as their hepatoprotective activities.From the title species Astragalus membranaceus(Fisch.)Bge.Var.Mongholicus(Bge.)Hisao,fourteen compounds,including five cycloartane triterpenoids,i.e.Astragal oside ?(1).Astragaloside ?(2).Isoastragaloside ?(3)?Isoastragaloside ?(4)?A stragaloside ?(5),two lupane triterpenoids Lupenone(6)and Betulin(7),four flavonoi ds compounds Methylnissolin(8),Isomucronulatol7-glueosid(9),7,3'-diohydroxy-5'-m ethoxisofiavone(10)and Calysosin-?-D-glucoside(12),two steroids compounds ?-sito sterol(13)and p-daucosterol(14),and one amide compound(2S,3S,4R,10E)-2-[(2R)-2-hydroxytetracosanoylamino]-10-octadecene-1,3,4-triol(11),were isolated by a combi nation of silica gel,macroporous resin,Sephadex LH-20,and C18 reverse-phased chrom atographic methods.Morever,eleven compounds,including three new seco-cycloartane triterpenoids Dikamaliartane G(15),Dikamaliartane H(16),Dikamaliartane ?(17),one known seco-cycloartane triterpenoids Dikamaliartane D(18),two known cycloartane triterpenoids Gardenolic acid A(19)and Gardenolic acid B(20),three known pentacyclic triterpenic acid Uosolic Acid(21),Rotundic acid(22),3?-6?-23-trihydroxyolean-12-en-28-oic acid(23),two iridoid compounds Aglucone(24)and Cyclopenta(25),were isolated from the 95%EtOH extract of the flower of Gardenia jasminoides Ellis by a variety of chromatography techniques,including silica gel,D101 macroporous resin,Sephadex LH-20,and Cix reverse-phased silica gel chromatography.The isolated compounds were elucidated on the basis of UV,IR,CD,MS,1D and 2D NMR spectroscopic techniques,among which three compounds were identified as new seco-cycloartane triterpenoids.Furthermore,the hepatoprotective activities of the characteristic triterpenes were evaluated on H2O2 induced cell damage model in HepG2 cells,and some compounds showed potential liver-protective activity.The above findings provided scientific reference for further studies on the two traditional Chinese medicine in health food raw materials.
