Studies On Exteraction And Purification Of DT-19 And Metabolism Of Gardenia Jasminoides Ellis

Gardenia jasminoides Ellis was early recorded in the book named" Sheng Nong's herbal classic",which is dry ripe fruit of Rubiaceae plant.Its function is to purge intense heat,remove annoyance,clear heat and promote diuresis,cool blood and detoxicate.DT-19 is the main compound of Iridoid compounds in Gardenia jasminoides Ellis.The documents records it can dramatically relieve cardiac failure.The essay measures and compares the DT-19 contents of ten batches Gardenia jasminoides Ellis from different producing areas.The research concludes the methods that the purification of DT-19 is extracted from medicinal materials.After animals take DT-19,the LC-ESI-Q-TOF-MS/MS is applied to the analysis and identification of the animals' metabolite.The main results are obtained as follows:(1)The research establishes the method to measure the HPLC-UV contents of DT-19 in Gardenia jasminoides Ellis.linear equation is y=703.29x+209.1 5 R=0.9996.The method RSD is 0.31%,the stability RSD is 0,55%,repetition RSD is 0.59%,average sample recovery rate is 97.75%,and RSD is 0.62%.It indicates the method is reliable and stable.By comparing the DT-19 contents of ten batches Gardenia jasminoides Ellis from different producing areas,it manifests the medicinal materials with highest DT-19 contents which is 0.69%are produced in Jiang Xi province.(2)The research concludes the methods that the purification of DT-19 is extracted from medicinal materials.By investigating solvent filtering,single factor test,and orthogonal test,the extraction of DT-19 is reflux extraction of 40%ethyl alcohol,with two hours and 3 times.The ratio of extracted liquid is 12:1.After extractive with DT-19 is separated and purified through macroporous resin and high performance liquid chromatograph,high purified samples of monomer is obtained.(3)After the rat takes DT-19,the technology of LC-ESI-Q-TOF-MS/MS is applied to analyze systematically the body composition(prototype components and metabolite)of portal blood,the whole body blood,urine,faces,heart,liver,spleen,lung and kidney.Prototype components and three metabolites are found in portal blood;Prototype components and four metabolites are found in the whole body blood;Prototype components and seven metabolites are found in the urine;Prototype components and two metabolites are found in the faces;Prototype components and two metabolites separately in heart,liver,spleen,lung and kidney.The analyzed results preliminarily demonstrate the distribution of prototypes and metabolites measured after the animal takes DT-19 in the body.The research provides the experimental basis for the relation between medicinal efficacy and organism.
