Objective : To study mechanism of renal protective effect of Mongolian medicine Chagan sugmel through animal experiment.Method : 100 Wistar rats(180~220g)were randomly divided to into 5 groups: normal group,model group,Chagan sugmel low dose group,Chagan sugmel high dose group and positive control group(Liu Wei Di Huang Wan).Begining from fifth day of experiment,by intraperitoneal injection of 100mg/kg gentamicin sulfate induced acute renal failure in rats control group and each treatment group for 5 days,continuously Chagan sugmel to administered in rats of Chagan sugmels two group and saline solution to administered in rats of normal group and model group for 21 days.On 21 st day to measured urine of every rat and to determined 10 indicator in urine.Also to observed such as emerged changes on body of rat,body weight changes of rat and death quantity of rat.At the end of the threatment period took blood from abdominal aortic and to separated serum.The by biochemical analyzer determined contents of BUN,Cre(Creatinine),ALB(albumin),TG(Triglycerides)and TC(Cholesterol)in serum of rat.The to took left kidney of rat in each group,to measured renal length and renal index and to observed pathological changes of renal tissue on light microscopy.Effect of endogenous substances analysed by methods with UPLC-MS(Ultra performance liquid chromatography-mass spectrometry),PCA(principal component analysis)and OPLS-DA(orthogonal partial least squares discriminant analysis)technology.Result : After the treathment of Chagan sugmel,body weight of rat in normal group compare to body weight of rat in other group,from 10 th day of Chagan sugmel treathment was decreased body weight of rat in model group and was increased body weight of rat in each Chagan sugmel group.Experimental rats were respectively died 0% of normal group,60% of control group and Chagan sugmel low dose group,65% of Chagan sugmel high dose group and 25% of positive control group.When control group compare to normal group,contents of ALB,BUN and Cre of serum and GLU and KET of urine in control group was increased(p<0.05)compare to normal group.But when Chagan sugmel high dose group compare to control group,renal index and renal length in Chagan sugmel high dose group was decreased(p<0.05)compare to control group.Also contents of GLU and KET of urine and ALB,BUN and Cre of serum in Chagan sugmel high dose group was significantly decreased(p<0.01,p<0.05)compare to control group and pathological changes in Chagan sugmel high dose group was decreased.The by analysis of PCA and OPLS-DA found 29 kind of endogenous biomarker in normal group and treatment groups.That's 29 kind of endogenous biomarker was significantly increased(p < 0.01)in Chagan sugmels two group.Conclusion: Mongolian medicine Chagan sugmel has renal protective effect against gentamycin sulfate induced renal failure in rats,especially Chagan sugmel high dose group has renal protective high effect.In PCA and OPLS-DA found 29 kind of endogenous biomarker and sphingosine,plant sphingosine sphingomyelin and arachidonic acid etc biomarkers are believed significantly endogenous biomarkers.That's significantly endogenous biomarkers maybe fundamental cause of renal protective effect of Mongolian medicine Chagan sugmel.
