Effect Of Ziyin Huoxue Jiedu Decoction On Thrombin Toxic Injury Of Focal Cerebral Infarction Rats

Objective:To observe the influence of Ziyin Huoxue Jiedu decoction (ZHJD) on the rats'cerebral infarct volume, The expression of prothrombin mRNA and PAR-1 mRNA of brain tissue and the contents of IL-1β,IL-8,TNF-αin serum after focal cerebral infarction,as well as to explore neuroprotective effects of Ziyin Huoxue Jiedu decoction.Methods:The 60 healthy SD rats were selected. The rats were randomly divided into 4 groups:the sham-operated,ischemia, ischemia with Ziyin Huoxue Jiedu decoction (ZHJD), ischemia with Argatroban Injection(AI),Each group was divided into three time phase point of lday, 3days,7days again,5 rats in each group. The model was reproduced by occluded middle cerebral artery. The expression of prothrombin mRNA and PAR-1 mRNA of brain tissue were detected by reverse transcription-polymerase chain reaction (RT-PCR) technique.The cerebral infarct range by TTC;The content of IL-1β,IL-8,TNF-αin serum were measured by enzyme—linked immunosorbent assay (ELISA).The results show:1. After cerebral ischemia, brain has no infarction in the sham group; after 7 days,the model group,the ZHJD group,the AI group were visible infarction. Infarction volume analysis shows that:Infarct volum ratio in the ZHJD group and AI group were all significant lower than the model group (P<0.01),while infarction volume percentage in the ZHJD group was obviously lower than the AI group(P<0.05);2. After the ischemia,the expression of prothrombin mRNA level as cerebral ischemia time extended and increase. Compared with sham group, the expression of prothrombin mRNA of brain tissue of the model group,the ZHJD group and the AI group were increased obviously (P<0.01); Compared with model group, the expression of prothrombin mRNA of brain tissue of the ZHJD group and the AI group were significantly decreased (P<0.01); The expression of prothrombin mRNA of two time phase point of the ZHJD group was significantly higher than the AI group (P<0.01).3. After cerebral ischemia of 1 days and 3days, compared with the sham group, the expression of PAR-1 mRNA of the model group was significantly increased (P<0.01);AI group in the treatment of 1 days and 3 days,the expression of PAR-1 mRNA were significantly lower than model group;The ZHJD group in the treatment of 1days,the expression of PAR-1 mRNA was not decrease significantly(P>0.05),after the treatment of 3 days, that was decrease significantly (P<0.01),but the decline was significantly lower than the AI group (P<0.01).4.Cerebral ischemia from 1 to 7 days, The content of IL-1βin serum was remained at a high level state,compared with the sham group at the same time (P<0.01).ZHJD and Argatroban can significantly reduced the increase of IL-1βcaused by ischemia (P<0.01),the inhibitory effect was time dependence (P<0.01);Treatment 1 day, The effect of ZHJD to IL-1βwas significantly better than Argatroban (P<0.01),but 3,7 days of treatment, Argatroban's inhibition of IL-1βwas obviously better than ZHJD (P<0.01).5. Cerebral ischemia from 1 to 7 days, The content of IL-8 in serum was remained at a high level state,compared with the sham group at the same time (P<0.01).ZHJD and Argatroban can significantly reduced the increase of IL-8 caused by ischemia (P<0.01),the inhibition was time dependence (P<0.01); The inhibitory effect of ZHJD and Argatroban to IL-8 was consistent (P>0.05)6. Cerebral ischemia from 1 to 7 days, The content of TNF-αin serum was remained at a high level state,compared with the sham group at the same time (P<0.01).ZHJD and Argatroban can obviously reduced the increase of TNF-a caused by ischemia (P<0.01),the inhibitory effect was time dependence (P<0.01); The inhibitory effect of ZHJD and Argatroban to TNF-αwas consistent (P>0.05)Conclusion:Zinyin HuoxueJiedu decoction is effective to restrain the expression of prothrombin mRNA and PAR-1 mRNA of brain tissue, reducing inflammation and cerebral infarct size,which may be one of cerebral protective effects of Ziyin Huoxue Jiedu decoction.
