Background and Objective Osteosarcoma is an primary idiopathic and highly malignant tumor which derived from mesenchymal tissue and common age of patients is about 20 years old of the following young men and children.Osteosarcoma is to metastasis in a early ago which leads to a high mortality clinically and seriously endangering the health of teenagers.The aim of current study is to investigate the protein expression of CD147,GLUT1 and MMP-9 and its clincopathological features and their relationship in osteosarcoma and osteochondroma.Methods(1)The protein expression of CD147,GLUT1 and MMP-9 was examined in 50 cases of osteosarcoma and 20 cases of osteochondroma tissue specimens by S-P immunohistochemical method.(2)The expression of CD147,GLUT1 and MMP-9 m RNA was detected by real-time PCR assay after transfection of CD147 si RNA and control si RNA in MG-63 cells by Lipofectamine.Results(1)The positive rates of CD147,GLUT1 and MMP-9 protein were 54.5%?67.3%and 72.2% in osteosarcoma respectively,which were significantly higher than those in osteochondroma(All P<0.05).The expression of CD147 was significantly correlated to tumor soft tissue invasion and Ennecking stage(both P<0.05).The expression of GLUT1 was significantly associated with tumor size and MMP-9 expression was significantly associated with male patient,tumor size,tumor soft tissue invasion,tumor grade and Ennecking stage(All P<0.05).Furthermore,the expression of CD147 was positively correlated to both GLUT1 and MMP-9 expression in osteosarcoma(r=0.568 and 0.220,both P<0.05).(2)Expression of CD147,GLUT1 and MMP-9 m RNA was significantly decreased after transfection of CD147 si RNA,compared with them transtection with control si RNA(All P<0.01).Conclusion CD147 might play an important role in the glucose metabolism,invasion and metastasis of human osteosarcoma by regulating the expression of GLUT1 and MMP-9respectively.
