Objective:The selection of three different time of moxibustion treatment of experimental knee osteoarthritis rabbit. Aims to observe the synovial fluid of IL-1, TNF-a content and articular cartilage type ? collagen expression and their mRNA expression of the relationship between changes and to explore and moxibustion in the treatment of KOA moxibustion, effect, moxibustion KOA choose appropriate moxibustion long provided experimental basis.Methods:50 Japanese White Rabbits, male and female half, weight 2.5±0.25 kg. The experimental animals were randomly divided into control group, model group, 10min treatment group,20min treatment group and 40min treatment group,10 rats in each group. Then the model group, 10min treatment group,20min treatment group and 40min treatment group were used in the experimental rabbits, the right knee was injected into the cavity of the KOA model. The blank group and normal saline was injected into the right knee 0.5ml. After the model was made, the 3 treatment groups were treated with "Wai xiyan" and "Nei xiyan".10min treatment group,20min treatment group and 40min treatment group were treated once a day,5 times for a course of treatment, the interval between 2 days, a total of 4 courses of treatment.After the end of treatment from each animal right knee joint femoral medial condyle cartilage and synovial membrane and surrounding tissue, with HE staining and Masson staining in pathological analysis and immunohistochemical method was used to determine the content of type II collagen, with real-time fluorescence quantitative polymerase chain reaction (RT-PCR) method to detect the type II collagen mRNA expression; selected animals in each group of the right knee joint synovial fluid, the content of ELISA detection of IL-1? and TNF-?. The income data are used spss21.0 were statistically analyzed, using single factor analysis of variance was used for statistical analysis, between the two groups were compared using LSD, data are used "mean±standard deviation" to represent, P< 0.05 said the difference was statistically significant.Results:1.HE staining results:compared with the control group, model group showed obvious inflammatory cell infiltration; compared with the model group, 10min,20min and 40min group obvious infiltration of inflammatory cells were significantly reduced; 10min,20min and 40min group compared to 40min group small amount of inflammatory cell infiltration of the specimens was significantly larger than that in the other two groups; 10min and 20min groups no significant difference.2.Masson staining absorbance value (OD):compared with the control group, the OD value of model group decreased significantly (P< 0.01); compared with the model group,1 Omin,20min and 40min group OD value increased significantly (P< 0.05); 10min,20min and 40min group compared, the OD value of the 40 Min group increased very significantly (P< 0.01); compared with the group at 10 min,20 min group no statistical difference (P> 0.05).3.Detection of IL-1 ? and TNF-a in joint fluid:compared with the control group, in model group, the articular fluid IL-1? and TNF-a increased significantly (P< 0.01); compared with the model group, lOmin,20min and 40min group joint fluid of IL-1? and TNF-a were significantly decreased (P< 0.01),10min,20min and 40min group compared,40min group decreased IL-1? and TNF-a levels than the other two groups more significant (P< 0.01).4. Type II collagen content and mRNA expression detection:compared with the control group, model group in type ? collagen content and mRNA expression decreased significantly (P< 0.01); compared with the model group, 10min,20min and 40min group in type II collagen content and its mRNA expression increased significantly (P< 0.05);10min,20min and 40min group compared 40min group of type II collagen content and its mRNA expression than the other two groups was more significant (P< 0.05); compared with the group at 10 min,20 min group no significant difference (P>0.05).Conclusion:1. This experiment used to the rabbit's knee joint cavity injection of the solution of the papain solution in order to successfully reproduce the experimental KOA model.2. In the development of pathology of KOA, synovial inflammation infiltration is to accelerate cartilage matrix decomposition, destruction of articular cartilage.3. 10min,20min and 40min Moxibustion "Nei xiyan", "Wai xiyan"point, by reducing the experimental knee osteoarthritis synovial fluid IL-1?, TNF-? to control synovial inflammation infiltration, improve type II collagen expression and mRNA expression level and delay the degeneration of articular cartilage.40min group is the most significant.
