Gastric cancer is a complex disease and it is the third in the world, after lung cancer and breast cancer. Although with the advancement in technology, the treatment of this disease has been some improved, but because the incipient symptom is unconspicuous resuling in that 5-year survival rate of this disease is only 20%-30%. For increasing the level of incipient diagnosis and precaution, it is more important to find some dependable molecular markers correlated with gastric carcinogenesis. Particularly the Wnt signaling pathyway plays an important part in carcinogenesis and the aberrant expression of some antagonistic genes will activate the pathyway leading to carcinogenesis.Therefore in this experiment we mianly disscuss the expression level of antagonistic genes(DKK1?DKK2?DKK3?SFRP2?SFRP2?SFRP5?APC?AXIN2) in the Wnt signaling pathyway and the correlation between expression level and clinicopathological parameters. Our aim is to find molecular markers for the incipient diagnosis and clinical assessment.We all collected 40 cases gastric cancer and corresponding adjacent frozen fresh tissues, then use RT-PCR Taqman probe method to detect the expression level of the antagonistic genes and use 2-? ct or 2-?? ct formula to process the exoression data. We mainly draw the following conclusions:a. Using Mann-Whitney nonparametric rank sum test to compare the 2-? ct value, we found DKK2 (P<0.0001), SFRP2 (P= 0.049), SFRP5 (P= 0.004) gene relative expression level were significantly lower in gastric cancer and DKK1, DKK3, SFRP4, AXIN2, APC were no significant difference, indicating the down-regulated expression of DKK2, SFRP2, SFRP5 plays an important part in the development of gastric carcinogenesis; b. Using Mann-Whitney nonparametric rank sum test to compare the 2-?? ct value, we also found that DKK2, SFRP2, SFRP5 were significantly lowered, illustrating these three genes may play a more important part in the pathway; c. Using t tese to analysis expression data and clinicalpathological data, we found that all genes were no significant difference with age, gender and differentiation; the expression level of DKK1, DKK2, DKK3, SFRP2, SFRP4, APC were significantly associated with TNM stage;and the abnormal expression of all genes were also statistically significant with lymph node metastasis.In conclusion, the dysregulated expression of the inhibiting gene plays a more important role in the gastric carcinogenesis in Wnt signaling pathway, and the down-regulated expression of related genes are closely associated with the occurrence and progression of gastric cancer, which may be used as molecular markers of gastric early diagnosis.
