Objective:By using liquid chromatography mass spectrometry(LC- MC) for gastric cancer patients and healthy people blood testing, to find out the difference of metabolic map between gastric cancer patients and healthy people. According to the difference of metabolic map, the patients with gastric cancer were divided into groups, then analyse the difference of clinical pathological parameters between the groups. And finally search for metabolites that can be used to analyze the prognosis of patients with gastric cancer.Methods:1. Sample preparation: The serum samples were thawed at room temperature,then adding acetonitrile. Vortex, centrifuge and suck the supernatant to reserve.2. Chromatography mass spectrometry analysis: samples were performed gas chromatography-mass spectrometry analyzer for metabolic products, then further statistical analysis of collected metabolic patterns,summarize the experimental data.3. Data analysis: Using t test, principal component analysis,PLS-DA,cluster analysis, variance analysis, COX proportional hazard regression analysis and other statistical methods to analyze the experimental data.Analysis of metabolic profiles of gastric cancer patients and healthy individuals, as well as the difference of clinical pathological parameters of gastric cancer patients with different metabolic maps. And to search for metabolites that can be used to analyze the prognosis of patients with gastric cancer.Results:The results of the PCA score map of the metabolites of blood samples showed that patients with gastric cancer and healthy people can be well differentiated. There are 87 kinds of metabolites in gastric cancer patients and healthy people by t test. Then through the principal component analysis, discriminant analysis and other methods, 16 metabolites were found can be used in the differentiation of gastric cancer patients and healthy people. According to the expression of the 16 metabolites in all patients with gastric cancer, the patients with gastric cancer were divided into groups.By comparing the results found, Each group there were significant differences in age, tumor stage, tumor differentiation, tumor location, gene expression and other clinical pathological parameters. Finally through the COX proportional hazard regression analysis,3 metabolites were associated with the prognosis of gastric cancer patients.Conclusion:By using the liquid chromatography mass spectrometry analysis of the blood of gastric cancer patients and healthy people, the differences of metabolites in serum and normal human serum were found. 16 metabolites were obtained, which can be a good distinction between gastric cancer patients and healthy people. The gastric cancer patients were divided into different groups according to the different metabolic products. There was a significant difference in the various clinical pathological parameters by comparing the groups, The close relationship between the serum metabolic group and the biological behavior of gastric cancer was proved. And 3 metabolites were found to be related to the prognosis of gastric cancer patients.
