| Objective: This study base on contrastive analyze the drug concentration that is measured by Micro-PET and Gamma counter to evaluate the application value of Micro-PET in dynamic monitoring the concentration of 18F-FDG in vivo.Methods: 1.Grouping and staging of the experiment: 10 rats were seen as experimental group and control group respectively by the self-contrast method.The experiment were divided into early(within 60 min)and late(after 60 min)according to the time after injecting of 18F-FDG.2.The experimental group : The rates of the experimental group were injected 18F-FDG,and were scanned by Micro-PET at the early experiment and the late experiment(included 90min、120min、240min、360min and 480min).The data were reconstructed by PIwork software,and gained the average dose of radiation at the different time points by sketching left ventricular cavity as region of interest(ROI),as well as calculated blood drug concentration Cexperiment of 18F-FDG.3.The control group : After 10 days(avoided experimental radioactive interference),The rates of the control group were injected the same dose of 18F-FDG,and got blood samples by cutting tail at 2 min,5 min,10 min,15 min,20 min,30 min,45 min,60 min,90 min,120 min,240 min,240 min,360 min and 480 min time point respectively.The count per minute of blood were determined quantitatively by Gamma counter,and calculated the blood drug concentration Ccontrol of 18F-FDG at different time points.4.SPSS 19.0 software was applied to statistical analysis and Excel software was applied to make the relevant diagram;The measurement data applied to x±s as a representation.Compare the experimental group and the control group of data by the paired sample t test.The Pearson correlation analysis,the within group consistency analysis and linear regression analysis were to the two groups of data of the early experiment,and Excel was applied to make the related chart.Results: 1.The early experiment: The two groups ofblood drug concentration at the early experiment was no statistical difference(P >0.05),and Pearson correlation coefficient r was 0.986(P=0.000),namely the two groups of data had remarkable correlation.Average measure ICC and 95%CI were0.976 [0.891,0.995],namely the two groups of data had remarkable consistency.The deterministic coefficient R2 was 0.984,namely the two groups of data had good linear relationship.In a word,the drug concentration of 18F-FDG which was measured by two methods of testing was same.2.The late experiment:The two groups of blood drug concentration at the late experiment was statistically different(P <0.05),namely the drug concentration of 18F-FDG which was measured by two methods of testing was different.Conclusion: Micro-PET could dynamic continuously monitor drug concentration of 18F-FDG at the early experiment,and can be a effective method to measure concentration of 18F-FDG in vivo. |
