Based on the multivariate spectrophotometry, the fast analysis of the complicated samples and the simultaneous determinations of multi-component blood concentration with dissociate and conjunct drugs are researched in the paper.The multivariate calibration models of the two drugs compound clorprenaline and compound paracetamol are established with the method of near infrared diffuse reflectance spectroscopy and principal component analysis—support vector machine regression (PCA-SVR). With the same principal components, the fitting and prediction capability of the models built by PCA-SVR is compared with the traditional robust method partial least squares regression (PLSR). The fast and simultaneous determination method for the effective components in the two compound drugs has been established.In the paper, three-dimensional fluorimetric and full rank parallel factor analysis is used in the determination of the levofloxacin blood concentration. First of all, the three-dimensional fluorescence spectra of levofloxacin in different pH values are acquired and the corresponding three-way array is used in the data resolution. Then the pure spectra of levofloxacin with different structures are obtained, of which one is HDS (hydrogen bond structure) and the other is DIS (dipole ion structure). With the pure spectra of HDS and DIS as the reference spectra, the conjunction component of levofloxacin and protein in the plasma samples is studied and the pure spectral of DIS-Protein component is obtained. Based on the results of the three pure spectra, the specification curves of DIS and HDS in plasma samples are established. The conclusion is obtained that the two specification curves could be used in the determinations of the blood concentration of levofloxacin by the results of recovery. With the help of specification curves, the concentration variation of DIS and HDS in the plasma samples of rabbit is studied and the inference is obtained that the variation trend of DIS and HDS in the samples is caused by the change of the rabbit internal environment. Meanwhile, the determination method for the blood concentration which have been established in the paper is credible. Finally, the blood concentration of levoflxacin in the whole blood samples is determined, the recovery and precision of the method is good.
