The MiRNA In Hp Infection Related MiRNA Is Associated With Hp Infection With Gastric Cancer And Clinical Research On The Characteristics Of Syndromes In Chinese Medicine

Objective: 1.This experiment aims to explore gene expression profile chip technology screening of gastric cancer and normal gastric tissue gene expression differences of feasibility.Then hope to be able to clear the infection of the mi RNA in the role of Hp in gastric cancer.2.This experiment used to explore the regular between the mechanism which one has the gastric cancer also infection by Hp and the distribution of TCM symptoms.Methods: 1.Using the Agilent 2100 Bioanalyzer gene chip detection the mi RNAs of the 6 patients who has the gastric cancer and the 6 cases of the normal gastric mucosa tissue.Then select expression the differences of mi RNAs.And using the mi RNA target gene prediction software to predict the top ten of the mi RNA target genes differentially expressed.And to use the GO and Pathway analysis to analyzed the target genes.2.To collect the patients who have gastric cancer after checking in hospital?the diagnostic criteria of Western medicine,the diagnostic criteria of traditional Chinese medicine,the diagnosis standard of the electronic endoscope,the diagnosis standard of Hp?and put them?the general information,gastroscope data,the pathological dignostie result,the four diagnostic data of TCM?in this study.For syndrome differentiation of traditional Chinese medicine to group the patients who have gastric cancer according to the related clinical data?abdominal pain,abdominal pain,diarrhea,heartburn,acid reflux,constipation,difficulty swallowing,nausea,vomiting,digestive hemorrhage and so on?,collecting data into the data statistical analysis,using SPSS16.0 count data expressed as a percentage of measurement data to or median,said rate compared with chi-square test,with p<0.05 for the difference was statistically significant.Results: 1.People gastric cancer tissue and normal gastric tissue group compared gene expression difference ratio?2.0 a total of 712 genes,including selected 10 down expression of mi RNAs?P-value?0.05?,the top 10 different mi RNA target genes are respectively: hsa-mi R-1296-5P,hsa-mi R-132-3P,hsa-mi R-155-5P,hsa-mi R-19-3Pb,hsa-mi R-19-3Pb,hsa-mi R-204-5P,hsa mi R-301-b-3P,hsa-mi R-a-3P30,hsa-mi R-3613-5P,hsa-mi R-625-3P,hsa-mi R-will-chr3₅397,clustering analysis,show the stomach group 1,4,5,and 6 samples with good consistency,gastric cancer samples of 2,3,more inclined to the normal group;Target gene Pathway analysis shows: the mi RNA target genes and the Hedgehog,MAPK,Estrogen,takes the cell carcinoma,Rap1,Endocytosis,TGF-beta,Axon guidance,Proteoglycans in cancer,Focal adhesion and other pathways are closely related.2.This study collected 60 patients with Hp infection positive gastric cancer,which has a number of smoking history in 21 cases,accounted for 35%,including male patients for 19 cases,compared with 31.66%,female patients accounted for in2 cases,3.33%;Always,a total of 35 cases of patients with a history of heavy drinking,accounted for 58.33%,male patients accounted for 29 cases among them,48.33%,female patients accounted for 6 cases,10.00%.Dietary factors:in this study,60 patients with gastric cancer have poor eating habits of 47 cases,accounted for 78.33%,of which 21 cases like eating spicy excitant food,like eating a high salt diet 14 cases,12 cases like eating pickled smoked foods.Social mental factors: the study of 60 patients with gastric cancer have long affected by the social environment work spirit function in 26 cases,accounting for 43.33%.On smoking,drinking,diet and mental factors in multiple factors unconditioned Logistic regression analysis,using Wald before entered to the fitting of the variables,the results found that smoking,drinking,dietary factors and social factors of P values are less than 0.05,there are significant differences.Gastric cancer patients with TCM syndrome type distribution mainly spleen deficiency cold syndrome,followed by liver stomach with card,phlegmy wet grinds to card,stasis poison knot inside,thermal injury stomach Yin.One over the age of 60 patients with gastric cancer is given priority to with taste xu han,40⁵9 patients with liver stomach not give priority to,under the age of 39 patients with phlegm wet grinds to give priority to.Conclusion: 1.the differences in expression of genes to be screened can be used to verify the mi RNA's relationship with gastric cancer.However the analysis of the mechanism still needs further validation.2.This study found that gender,age,and dietary factors and cancer of the stomach of TCM syndrome distribution have certain relevance.
