| Background:NOD-like receptors play critical roles in the activation and regulation of innate immune response.NLRP3 is known as a member of NLRs family,and the NLRP3 inflammasome is confirmed to associate with various diseases,such as asthma,inflammatory bowel disease,metabolic disorders,multiple sclerosis,as well as other auto-immune and auto-inflammatory diseases.However,the role of NLRP3 in the chronic rhinosinusitis with nasal polyps(CRSwNP)has not been explored.Methods:Twenty-five specimens of nasal polyps and 15 specimens of uncinate processes from chronic rhinosinusitis with nasal polyps in patients and 15 specimens of middle turbinate from patients with deviation of nasal septum were collected.The experiment is divided into three groups:control group,uncinate group,nasal polyp group.Western blot assay was employed for analyzing the expression of NLRP3.The IL-1β was detected using immunohistochemistry(IHC).and real-time polymerase chain reaction(RT-PCR).Production of lipopolysaccharide(LPS)induced IL-1β with/without NLRP3 inflammasome inhibitor was measured using enzyme linked immunosorbent assay(ELISA)and western blot in cultured dispersed nasal polyp cells(DNPCs).Results:NLRP3 show high expression in nasal polyps than control group(P<0.01).Expression of IL-1(3 was significantly higher in the nasal polyps of CRSwNP than control group(P<0.05).LPS induced IL-1β production was significantly suppressed by treatment of NLRP3 inflammsome inhibitor(P<0.05).Conclusion:NLRP3 inflammasome played a pivotal role in the pathogenesis of CRSwNP,and MCC950 is thus a potential therapeutic for NLRP3 inflammasome mediated inflammation in nasal polyps. |
