Breast cancer represents the most common form of cancer in women.With improvements in growth rates for breast cancer early detection over the past decades.In 2015,the report indicates that breast cancer is the first(25%)growth rates in American new disease,exceeding 5 million women died in it and accounting for 15%.In recent years,clincal application of medicine for breast cancer could promote the recoverability after surgery and reduce side effect of radiotherapy and chemotherapy.Therefore,it is significant for women of breast cancer to find out the medicine that curing breast cancer.Ajuga decumbens(HBG)and Poria cocos(FL),the traditional Chinese medicine,are the common,which has the effect of inhibiting tumor,resisting inflammation and reinforcing immunity.To investigate the effect of iridoids in Ajuga decumbens,triterpenes in Poria cocos and their combination on the metastasis of invasive breast cancer cells and related mechanisms.We conducted several assays including cell adhesion assay,scratch assay and transwell invasion assay.In vivo,detecting transfer of breast cancer cells in the body by the models of subcutaneous tumor,pulmonary and bone metastases.Western blot analysis was employed to detected the expression of associated proteins of EMT and MAPK signaling pathway.FL,HBG and their combination could significantly inhibited the cell adhesion,migration and invasion of MDA-MB-231 and SK-BR-3 cells.HBG and FL has the effect of inhibiting propagation tumor,and has the effect of reducing the bad results of pulmonary and bone metastases,which compatibility has good synergistic effect.The effect of HBG and FL has markedly synergistic effect,and the best compatibility ratio was 10:1.HBG,FL and their combination could reverse EMT of breast cancer cells,which increased the levels of epithelial biomarkers,such as ?-catenin,E-cadherin and ZO-1 and reduced the levels of mesenchymal biomarkers,such as vimentin.Moreover,treatment of the cells with HBG,FL and their combination resulted in markedly inhibition of phosphorylation of ERK1/2,JNK and p38.These results indicated that the combination of HBG and FL have the ability to inhibit breast cancer cell invasion,and do so by targeting the expression of MAPK pathway as well as suppressing the epithelial-to-mesenchymal transition.This experiment by studying the HBG,FL alone and combination of the effects of cell invasion and metastasis using assaies of molecular and vivo of carrier,clarifying its value of medicine to inhibit tumor,for itspotential clinical application in breast cancer care and to provide theoretical basis for compound compatibility theory to make a meaningful availability.
