Pumpkin from Pueraria,Pueraria and Pu'er tea polysaccharides,Pueraria flavonoids and tea brown powder because of its toxic side effects,is widely used as health food raw materials.However,there are few reports on the effect of the three plant extracts on improving the interaction between type 2 diabetes mellitus and the mechanism of the three kinds of plant extracts.Causing the cause of diabetic disease complex,the body's glycolipid metabolism system and the distribution of intestinal flora and many protein pathways show abnormal.In this paper,the animal model of type 2 diabetes mellitus was established to study the effect and mechanism of single and combined use of three kinds of plant extracts on metabolic disorders of type 2 diabetes mellitus.In this paper,the type 2 diabetes model was established and the Optimal dose of the pumpkin polysaccharide,pueraria flavonoids and pu-erh theabrowns were determined by measured blood glucose.On the basis,the fasting blood glucose(FBG),insulin(INS),insulin resistance(IR),high density lipoprotein(HDL)and low density lipoprotein(LDL)were also investigated.The FBG,INS,IR,HDL,LDL combined group was significantly better than the single component(P < 0.05).The results revealed that the combination of pumpkin polysaccharide and pu-erh theabrowns,pueraria flavonoids and pu-erh theabrowns,pumpkin polysaccharide factorial analysis showed that the interactions of the three substances were interactive.The synergistic experiment showed that pumpkin polysaccharide combined with Pueraria lobata flavonoids or combination with tea-flavonoid significantly reduced TG content(P < 0.05).The combination of theaflavin and pueraria flavonoids in reducing the content of FBG,TC,LDL was significantly better than flavonoids,and the effect of TG and INS was better than that of thepueraria flavonoids and pu-erh theabrowns group(P < 0.05).The molecular mechanism experiments showed that the three components of a single component and two or two in the expression of PI3 K / Akt protein expression have enhanced role.Intestinal flora experiments showed that the type 2 diabetes mellitus intestinal flora disturbance,beneficial bacteria to reduce the increase in harmful bacteria,three kinds of plant extracts combined to make the mouse intestinal flora closer to normal,the best regulation.
