Objective:To compare the type 2 diabetic having no complications or having earlier period change of renal function with health adult on the difference of clinical index and intestinal flora, and analyzing the effect of intestinal flora on the correlated clinical index, and exploring the effect of intestinal flora on the developement and generate of diabetic nephropathy, and proved a new method for prevention and cure of diabetes.Methods:1.20 healthy volunteers?23 patients with type 2 diabetes having no complications and 29 patients with type 2 diabetes having earlier period change of renal function were elected, all of them were 45-65 years old.2.To gather the general numerical data and to detect the clinical data, such as the blood and urine for three groups of researched crowd, but the dejecta was stored for further researching.3.To compare general numerical data and clinical data among three groups of researched crowd, and find the correlated Risk factor of the developement and generate of diabetic nephropathy.4.The diversity of microbial population of dejecta in three groups of samples were detected with PCR-DGGE.The gel strips, which were conspicuous, were obtained and undertaked gene sequencing. The categories of conspicuous microbial population were confirmed after the sequencing results were contrast with GenBank.Results:1.The age?contractive pressure(SBP)?body mass index(BMI)?blood-fasting sugar(BFS)?triglyeride(TG)?alpha-lipoprotein(HDL)were statistically significantly different between type 2 diabetes and control group(healthy crowd); There are statistical differences(P<0.05).The age?course of diabetes?contractive pressure(SBP)?platelet/Homeocyte?glycosylated hemoglobin?empty stomach C peptid?uric acid were statistically significantly different between type 2 diabetes having no complications and type 2 diabetes having earlier period change of renal function; There are statistical differences(P<0.05).2.The developement and generate of type 2 diabetic nephropathy were positively correlation with age ? contractive pressure(SBP)? course of diabetes ?platelet/Homeocyte?glycosylated hemoglobin?uric acid, and were negative correlation with empty stomach C peptid, with Spearman correlated analysis(P<0.05).3.The course of diabetes?contractive pressure(SBP)and uric acid of type 2 diabetes were independent risk factors of developement and generate of diabetic nephropathy, with Logistic multiple regression analysis.4.The differences in number?position and brightness of straps among three groups atlas were found with PCR-DGGE finger printing analysis, which showed that there are differences in diversity of finger printing of intestinal flora among three groups.,and that there are high resemblance in every group intestinal flora, and the contrast of resemblance was conspicuous between groups. The abundance and band number of diabetic intestinal flora were decrease obviously compared with control group(healthy crowd)(P<0.05).There are differences in ascendancy bacterium among three groups, after gel extraction and gene sequencing have been finished from five conspicuous strap, the five corresponding bacterium, namely Prevotella? Eubacterium?Bifidobacterium?Flavonifractor plautii?Lactobacillus were identified.Conclusion:1.There are conspicuous difference in general numerical data and the clinical detect index among the healthy adult ? patients with type 2 diabetes having no complications and patients with type 2 diabetes having earlier period change of renal function. The course of diabetes?contractive pressure(SBP)and uric acid were independent risk factors of developement and generate of diabetic nephropathy.2.There are conspicuous difference in intestinal flora among the healthy adult?patients with type 2 diabetes having no complications and patients with type 2 diabetes having earlier period change of renal function.3.The difference of intestinal flora could be result in the changes of clinical detect index, and taking part in the generation of diabetic nephropathy.
