Objective To investigate the effect and mechanism of total flavonoids of Pueraria lobate(TFF)on retinal ganglion cells(RGC)in diabetic rats.Methods The diabetic model was induced by streptozotocin(STZ)in 60 male SD rats.Diabetic rats were randomly divided into three groups: diabetic group(diabetes mellitus,DM),TFF group(200 mg/kg)and metformin group(met,75mg/kg,positive control).15 normal rats were taken as control group(CON).After 12 weeks,Blood glucose,malondialdehyde(MDA)and superoxide dismutase(SOD)were measured,the number of RGC was detected by HE staining,the expression of Keap1,Nrf2 and HO-1 were detected by immunohistochemistry and Western blot.Results Compared with CON group,blood glucose,MDA content,the expression of Keap1 and Nrf2 were significantly increased,SOD activity and the expression of HO-1 were significantly decreased in DM group.Compared with the DM group,the blood glucose,MDA content,and Keap1 of the TFF group and the Met group were significantly reduced,and the SOD activity and the expression of Nrf2 and HO-1 were significantly increased.Conclusion TFF can protect RGC damage in diabetic rats by reducing blood glucose and enhancing the antioxidant capacity of retina,which be related to the regulation of Keap1-Nrf2/HO-1 signaling pathway.
