Objective In order to seek the best intervention time,the effects of beef decoction were investigated in neuroprotective effect of rats which suffered from cerebral ischemia injury at each time point.In addition to,effects of beef decoction and carnosine were observed on body weight,neurological function score,proinflammatory cytokine expression and infarct volume fraction in rats of cerebral ischemia to explore effect of beef decoction on Inflammation mechanism and optimal intervention time as well.Methods Male SD rats were randomly divided into sham group,model group,beef decoction group and carnosine groups,which are divided into 1 d,3 d and 7 d three subgroups,each subgroup have 8 rats.Prepared by suture permanent middle cerebral artery occlusion(pMCAO),beef decoction group was given by lavage with 600 mg·kg-1·d-1 beef decoction after 2 h,carnosine group with 600 mg·kg-1·d-1 camosine,sham-operated group and model group with the same amount of sterile saline 2 times a day.SD rats were killed and peripheral blood was isolated at the respectively corresponding time points.At each time point,after molding,body weigh was weighted by a scale;the neurological function was scored by Longa method;infarct volume fraction was revealed by 2,3,5-triphenyl tetrazolium chloride(TTC)staining,expressions of IL-1?,TNF-?,IFN-? and IL-10 were detected by protein enzyme-linked immunosorbent assay(ELISA).Results1.There was no significant difference in body weight in each group before pMCAO(P>0.05).After Id and 3d,body weigh decreased obviously on model group,beef decoction group and carnosine groups(P<0.01,P<0.05),but there were also no significant difference between the groups(P>0.05).After 7d,body weigh decreased obviously on model group,beef decoction group and carnosine groups(P<0.01,P<0.05).Compared with the model group,the body weight of the special beef decoction group was significantly higher(P<0.05).2.There was no significant difference in neurological function scores between groups at each time point after 2h(P>0.05).After 1d and 3d,compared with the model group,there was no significant difference in the neurological function scores between the beef decoction group and the carnosine group(P>0.05).After 7d,neurological function scores of beef decoction group and the carnosine group were reduced(P<0.01,P<0.05),and there was no significant difference between the two groups(P>0.05).3.After 3d,the volume fraction in beef decoction group and carnosine group increased,but there was no significant difference(P>0.05).After 7d,beef decoction group on the volume was significantly decreased(P<0.01).After 7d,carnosine group on the volume fraction decreased but there was no significant difference(P>0.05).4.After pMCAO,the levels of IL-1?,TNF-?,IFN-1? and IL-10 on the model group were significantly increased(P<0.05 or P<0.01).Compared with the model group,The levels of IL-1?,TNF-?,IFN-? and IL-10 were significantly decreased after 7 d the beef decoction soup(P<0.05).The expression of IL-1? was decreased after 7d carnosine,but the difference was not statistically significant(P>0.05).The levels of TNF-?,IL-10 and IFN-? were significantly decreased(P<0.01).5.The correlation coefficient of IL-1?,IFN-1?,TNF-?,TNF-?/IL-10 were r = 0.946,r=0.867,r=-0.852,r=-0.884,suggesting that there was a significant correlation between the volume of cerebral infarction and IL-1?,IFN-?,TNF-?,ratio of TNF-?/IL-10.The correlation coefficient was r = 0.194,suggesting that there was no significant correlation between cerebral infarction volume and IL-10 level(P>0.05).Conclusion1.600mg · kg-1 beef decoction is given rats of focal cerebral ischemia at least 7 days,which can increase the body weight of cerebral ischemia rats,significantly improve neurological deficits,reduce the volume of cerebral infarction and effect neuroprotective effects.2.600mg · kg-1 beef decoction can reduce the expressions of inflammatory cytokines(IL-1?,TNF-? and IFN-?)in rats with cerebral ischemia and increase the expression of inflammatory cytokines(IL-10).Significantly inhibiting inflammatory injury after cerebral ischemia.3.Compared with carnosine,the neuroprotective effect of the beef decoction with the same concentration of carnosine(600mg · kg-1)on cerebral ischemia was not statistically different.4.Beef decoction soup can be included as a simple diet side of the diet management of cerebral ischemia,which plays a supporting role on treatment of patients with cerebral ischemia.
