Effect Of Different Doses Of Beef Soup On Inflammatory Cytokines In Peripheral Blood Of Rats With Cerebral Ischemia

Abstract/Summary:
Objective This study intends to evaluate the effect of different doses of beef soup on inflammatory cytokines in peripheral blood of rats with cerebral ischemia,and to explore the relation between brain injury and inflammatory cytokines in order to investigate the mechanism of beef soup which protects cerebral ischemia from inflammatory injury,seeking the best intervention dose,in order to provide the evidence for clinical treatment of cerebral ischemia and theoretical guidance of diet for clinical cerebral ischemia patients.Methods 60 male SD rats(SPF scale)were randomly divided into six groups:the sham-operated group Ⅰ,the sham-operated group Ⅱ,the vehicle group,the low dosage beef soup-treated group(200 mg/(kg·d)),the middle dosage beef soup-treated group(400 mg/(kg·d))and the high dosage beef soup-treated group(600 mg/(kg·d).Permanent middle cerebral artery occlusion was established by suture method in the vehicle group and each beef soup groups.After vehicleing,the rats in the low,middle and high dose beef soup groups were lavaged with beef soup at the dose of 200,400,600 mg/(kg·d),respectively,the rats in the sham-operated group Ⅱ group were lavaged with beef soup of 600 mg/(kg·d),and the sham-operated group Ⅰand the vehicle group was lavaged with the same volume of normal saline.Before surgery and at the 7th day after surgery,the body weight of rats was observed,and at 2 h and 7 d after operation,the neurologic impairment was assessed with Zea Longa scale,The infarct volume fractions were measured by 2,3,5-triphenyltetrazolium chloride stain at 7 d after operation.The peripheral blood cytokines(TNF-α,IFN-γ,IL-6 and IL-4)were detected by ELISA at 7 d after surgery.Results1.Before the operation,the body weight of rats in each group were not significantly different(P>0.05),the neurological deficit scores of the beef soup-treated groups were also not significantly different at baseline(P>0.05).There were no significant difference about body weight,neurological function score,cerebral infarction volume and serum inflammatory cytokine expression between the sham-operated group Ⅰ and Ⅱ(P>0.05).2.Compared with the sham-operated group Ⅰ,the body weight of the vehicle group and the three beef soup-treated groups were significantly decreased at 7 d after surgery(all P<0.01),and compared with the vehicle group,the body weight had a rising trend after low and high dosage beef soup treatment(P>0.05).3.Compared with the sham-operated group Ⅰ,the neurologic impairment score in the vehicle group and the three beef soup-treated groups were significantly increased at 7 d after surgery(all P<0.01),and compared with the vehicle group,the neurologic impairment score had a significant decrease after high dosage beef soup treatment(P<0.01).4.At 7 d after operation,infarction were not observed in the brain tissues of rats in sham I group and sham II group,but were observed in the vehicle group and each beef soup group.The infarct volume fractions significantly decreased in the high dose beef soup groups compared with the vehicle group(P<0.05).5.The levels of serum IL-6,TNF-β,IFN-γ and IL-4 in the vehicle group were significantly higher than the levels in the sham I group(P<0.01,P<0.01,P<0.01,P<0.05).Compared with the vehicle group,the level of IL-6 decreased significantly(P<0.01),the levels of IL-4 and IL-4/IFN-γ ratio increased in the high dose beef soup group(P<0.01).The level of TNF-α significant decresased in the high and moderate dose beef soup group(P<0.01).The three dose beef soup could decrease the expression of serum IFN-y significantly(P<0.05,P<0.05,P<0.01).6.The infarct volume had positive correlation with the levels of serum IL-6 and IFN-y(r=0.607,P=0.002;r=0.696,P=0.000)and had negative correlation with the levels of serum IL-4(r=-0.453,P=0.026).The infarct volume had no correlation with the level of TNF-α(r=0.241,P=0.257).Conclusion1.beef soup can reduce weight losing and infarct volume of rats with cerebral ischemia,and improve scores of neurological deficits scales.2.beef soup can decrease the level of pro-inflammatory cytokines(IL-6,TNF-α and IFN-y)and increase the expression of anti-inflammatory cytokine(IL-4)in peripheral blood,thus inhibiting the inflammatory reaction after cerebral ischemia of rats.3.The infarct volume has positive correlation with the levels of serum IL-6 and IFN-y,and has negative correlation with the level of serum IL-4.It indicated that high expression of IL-6 and and IFN-γ in peripheral blood promote injury of brain,in contrast,high expression of IL-4 peripheral blood limit the infract volume and play a protective role in cerebral ischemia.
